The blood enzyme glutamate-oxaloacetate transaminase (GOT) has been postulated as an effective therapeutic to protect the brain during stroke. To demonstrate its potential clinical utility, a new human recombinant form of GOT (rGOT) was produced for medical use. We tested the pharmacokinetics and evaluated the protective efficacy of rGOT in rodent and non-human primate models that reflected clinical stroke conditions.

Nitrogen trifluoride (NF) is a potent and long-lived greenhouse gas that is widely used in the manufacture of semiconductors, photovoltaic cells, and flat panel displays. Using atmospheric observations from eight monitoring stations from the Advanced Global Atmospheric Gases Experiment (AGAGE) and inverse modeling with a global 3-D atmospheric chemical transport model (GEOS-Chem), we quantify global and regional NF emission from 2015 to 2021. Low-cost sensors (LCSs) for particulate matter (PM) concentrations have attracted the interest of researchers, supplementing their efforts to quantify PM in higher spatiotemporal resolution. The precision of PM mass concentration measurements from PMS 5003 sensors has been widely documented, though limited information is available regarding their size selectivity and number concentration measurement accuracy. In this work, PMS 5003 sensors, along with a Federal Referral Methods (FRM) sampler (Grimm spectrometer), were deployed across three sites with different atmospheric profiles, an urban (Germanou) and a background (UPat) site in Patras (Greece), and a semi-arid site in Almería (Spain, PSA).

Pancreatic ductal adenocarcinoma (PDAC) is among the deadliest cancers worldwide, primarily due to its robust desmoplastic stroma and immunosuppressive tumor microenvironment (TME), which facilitate tumor progression and metastasis. In addition, fibrous tissue leads to sparse vasculature, high interstitial fluid pressure, and hypoxia, thereby hindering effective systemic drug delivery and immune cell infiltration. Thus, remodeling the TME to enhance tumor perfusion, increase drug retention, and reverse immunosuppression has become a key therapeutic strategy.

Microplastics (MPs) are ubiquitous in marine compartments, and their transboundary distribution favours the dispersion and accumulation of particles in ecosystems. This study investigated MP contamination in four coastal fish species (Haemulon squamipinna, Chaetodon ocellatus, Syacium micrurum, and Alphestes afer) from the southwestern Tropical Atlantic. An alkaline treatment was applied to extract MPs from the digestive tracts, and a Laser Direct Infrared (LDIR) system was used to identify polymers.

This study compares the performance and output of an electrochemical phospholipid membrane platform against respective in vitro cell-based toxicity testing methods using three toxicants of different biological action (chlorpromazine (CPZ), colchicine (COL) and methyl methanesulphonate (MMS)). Human cell lines from seven different tissues (lung, liver, kidney, placenta, intestine, immune system) were used to validate this physicochemical testing system. For the cell-based systems, the effective concentration at 50 % cell death (EC) values are calculated.

Genotoxicity testing for nanomaterials remains challenging as standard testing approaches require some adaptation, and further development of nano-specific OECD Test Guidelines (TGs) and Guidance Documents (GDs) are needed. However, the field of genotoxicology continues to progress and new approach methodologies (NAMs) are being developed that could provide relevant information on the range of mechanisms of genotoxic action that may be imparted by nanomaterials. There is a recognition of the need for implementation of new and/or adapted OECD TGs, new OECD GDs, and utilization of NAMs within a genotoxicity testing framework for nanomaterials.

The past six years have been marked by some of the most dramatic climatic events observed in the Antarctic region in recent history, commencing with the 2017 sea-ice extreme low. The Humpback Whale Sentinel Programme is a circum-polar biomonitoring program for long term surveillance of the Antarctic sea-ice ecosystem. It has previously signalled the extreme La Niña event of 2010/11, and it was therefore of interest to assess the capacity of existing biomonitoring measures under the program to detect the impacts of 2017 anomalous climatic events.

The single cell gel electrophoresis technique is based on the measurement of DNA migration in an electric field and enables to investigate via determination of DNA-damage the impact of foods and their constituents on the genetic stability. DNA-damage leads to adverse effects including cancer, neurodegenerative disorders and infertility. In the last 25 years approximately 90 human intervention trials have been published in which DNA-damage, formation of oxidized bases, alterations of the sensitivity towards reactive oxygen species and chemicals and of repair functions were investigated with this technique.

The Fenwei Plain is one of China's most polluted regions, with poor atmospheric dispersion conditions and an outdated energy structure. After implementing multiple policies in recent years, significant reductions in air pollutant concentrations were observed. In this study, based on the Lagrangian-Bayesian inversion framework FLEXINVERT, we constructed a variable resolution inversion system focusing on the Fenwei Plain and inferred the carbon monoxide (CO) emissions using in-situ atmospheric CO observations from April 2014 to March 2020.

As part of a large human biomonitoring study, we conducted occupational monitoring in a glass fibre factory in Slovakia. Shopfloor workers (n = 80), with a matched group of administrators in the same factory (n = 36), were monitored for exposure to glass fibres and to polycyclic aromatic hydrocarbons (PAHs). The impact of occupational exposure on chromosomal aberrations, DNA damage and DNA repair, immunomodulatory markers, and the role of nutritional and lifestyle factors, as well as the effect of polymorphisms in metabolic and DNA repair genes on genetic stability, were investigated.

Black carbon emitted by incomplete combustion of fossil fuels and biomass has a net warming effect in the atmosphere and reduces the albedo when deposited on ice and snow; accurate knowledge of past emissions is essential to quantify and model associated global climate forcing. Although bottom-up inventories provide historical Black Carbon emission estimates that are widely used in Earth System Models, they are poorly constrained by observations prior to the late 20th century. Here we use an objective inversion technique based on detailed atmospheric transport and deposition modeling to reconstruct 1850 to 2000 emissions from thirteen Northern Hemisphere ice-core records.

With the current possible presence of thousands of PFAS compounds in industrial emissions, there is an increasing need to assess the impacts of PFAS regulation of conventional PFAS on one hand and the exposure to emerging and yet unknown PFAS on the other. Today's analytical methodologies using targeted approaches are not sufficient to determine the complete suite of PFAS present. To evaluate the presence of unknown PFAS, we investigated in this study the occurrence of an extended range of target PFAS in various species from the marine and terrestrial Norwegian environment, in relation to the extractable organofluorine (EOF), which yields the total amount of organofluorine.

Omics-based technologies have enabled comprehensive characterization of our exposure to environmental chemicals (chemical exposome) as well as assessment of the corresponding biological responses at the molecular level (eg, metabolome, lipidome, proteome, and genome). By systematically measuring personal exposures and linking these stimuli to biological perturbations, researchers can determine specific chemical exposures of concern, identify mechanisms and biomarkers of toxicity, and design interventions to reduce exposures. However, further advancement of metabolomics and exposomics approaches is limited by a lack of standardization and approaches for assigning confidence to chemical annotations.
