Antimalarial drug discovery has until recently been driven by high-throughput phenotypic cellular screening, allowing millions of compounds to be assayed and delivering clinical drug candidates. In this review, we will focus on target-based approaches, describing recent advances in our understanding of druggable targets in the malaria parasite. Targeting multiple stages of the Plasmodium lifecycle, rather than just the clinically symptomatic asexual blood stage, has become a requirement for new antimalarial medicines, and we link pharmacological data clearly to the parasite stages to which it applies.

In this study, an eco-friendly procedure was established by vortex-assisted liquid-phase microextraction based on deep eutectic solvent (VA-LPME-DES) combined with graphite furnace atomic absorption spectroscopy (GFAAS). The performance of this method was demonstrated by the extraction and analysis of lead (Pb), cadmium (Cd), and mercury (Hg) in fish samples. The hydrophobic DES is considered as a green extractant (environmentally friendly and less toxic than common organic solvents) and is a suitable alternative to common toxic organic solvents and is made of l-menthol and ethylene glycol (EG) with a molar ratio of 1:1.

In the work that is being presented here, the effect of sample thickness and laser irradiance on the reduction of the signal-to-background ratio SBG and the plasma parameters, specifically electron temperature and electron density, is being investigated using back-reflection-enhanced laser-induced breakdown spectroscopy (BRELIBS). Copper and silver discs that had been highly polished were attached to the back surface of the glass target, and the Nd-YAG laser beam that was focused on the front surface of the target was tuned to its fundamental wavelength. The thicknesses of the transparent glass samples that were analysed were 1 mm, 3 mm, and 6 mm.

Background: Analyses of factors that determine quality of perinatal care consistently rely on clinical markers, while failing to assess experiential outcomes. Understanding how model of care and birth setting influence experiences of respect, autonomy, and decision making, is essential for comprehensive assessment of quality.
Methods: We examined responses (n = 1771) to an online cross-sectional national survey capturing experiences of perinatal care in the United States.

Several implantable long-acting (LA) delivery systems have been developed for sustained subcutaneous administration of tenofovir alafenamide (TAF), a potent and effective nucleotide reverse transcriptase inhibitor used for HIV pre-exposure prophylaxis (PrEP). LA platforms aim to address the lack of adherence to oral regimens, which has impaired PrEP efficacy. Despite extensive investigations in this field, tissue response to sustained subcutaneous TAF delivery remains to be elucidated as contrasting preclinical results have been reported in the literature.

Unlabelled: Automation of deceased donor referrals with standardized clinical triggers allows organ procurement organizations to be rapidly aware of medically eligible potential donors without the need for manual reporting and subjective decision-making of otherwise very busy hospital staff. In October 2018, 3 Texas hospitals (pilot hospitals) began using an automated referral system; our goal was to evaluate the impact of this system on eligible donor referral.
Methods: We studied ventilated referrals (n = 28 034) in a single organ procurement organization from January 2015 to March 2021.

Identifying how small molecules act to kill malaria parasites can lead to new "chemically validated" targets. By pressuring Plasmodium falciparum asexual blood stage parasites with three novel structurally-unrelated antimalarial compounds (MMV665924, MMV019719 and MMV897615), and performing whole-genome sequence analysis on resistant parasite lines, we identify multiple mutations in the P. falciparum acyl-CoA synthetase (ACS) genes PfACS10 (PF3D7_0525100, M300I, A268D/V, F427L) and PfACS11 (PF3D7_1238800, F387V, D648Y, and E668K).

Development of antimalarial compounds into clinical candidates remains costly and arduous without detailed knowledge of the target. As resistance increases and treatment options at various stages of disease are limited, it is critical to identify multistage drug targets that are readily interrogated in biochemical assays. Whole-genome sequencing of 18 parasite clones evolved using thienopyrimidine compounds with submicromolar, rapid-killing, pan-life cycle antiparasitic activity showed that all had acquired mutations in the cytoplasmic isoleucyl tRNA synthetase (cIRS).

Public health agencies' ability to protect health in the wake of COVID-19 largely depends on public trust. In February 2022 we conducted a first-of-its-kind nationally representative survey of 4,208 US adults to learn the public's reported reasons for trust in federal, state, and local public health agencies. Among respondents who expressed a "great deal" of trust, that trust was not related primarily to agencies' ability to control the spread of COVID-19 but, rather, to beliefs that those agencies made clear, science-based recommendations and provided protective resources.
