Synthesis of erythrodiol C-ring derivatives and their activity against Chlamydia trachomatis.

To develop new potential agents against Chlamydia trachomatis among oleanane type triterpenoids the synthesis, spectral and X-ray analysis as well as antimicrobial screening of C-12 oxygen and nitrogen derivatives of erythrodiol is presented. The reduction of methyl 3β-acetoxy-12-oxo-oleanoate with LiAlH led to isomeric erythrodiol 12β- and 12α-hydroxy-derivatives, their stereochemistry with respect to the position of hydroxyl-group at C-12 was determined based on the multiplets splitting patterns, the magnitude of the spin-spin interaction, and NOESY interactions. Methyl 3β-acetoxy-12-oxo-oleanoate was transformed to 12E-hydroxyimino- and 12E-methoxyimino-derivatives by the interaction with NHOH∙HCl or CHONH∙HCl, respectively.

Evaluation of A-azepano-triterpenoids and related derivatives as antimicrobial and antiviral agents.

A series of semisynthetic triterpenoids with A-ring azepano- and A-seco-fragments as well as hydrazido/hydrazono-substituents at C3 and C28 has been synthesized and evaluated for antimicrobial activity against key ESKAPE pathogens and DNA viruses (HSV-1, HCMV, HPV-11). It was found that azepanouvaol 8, 3-amino-3,4-seco-4(23)-en derivatives of uvaol 21 and glycyrrhetol-dien 22 as well as azepano-glycyrrhetol-tosylate 32 showed strong antimicrobial activities against MRSA with MIC ≤ 0.15 μM that exceeds the effect of antibiotic vancomycin.

Synthesis of C17-[5-methyl-1,3]-oxazoles by -propargylation of triterpenic acids and evaluation of their cytotoxic activity.

A series of unexpected triterpenic C17-[5-methyl-1,3]-oxazoles along with targeted -propargylamides was synthesized by an interaction of acid chlorides with propargylamine hydrochloride. We proposed that the formation of methyl oxazole passes through an alternative pathway by the participation of the terminal alkyne carbon atom and acid chloride intermediate with following intramolecular rearrangements. A Weakly Antiferromagnetically Coupled Biradical Combining Verdazyl with Nitronylnitroxide Units.

An antiferromagnetically (AFM) coupled biradical based on oxoverdazyl and nitronylnitroxide was synthesized in 46 % yield using Sonogashira coupling. The obtained heterobiradical evidenced distinct properties of both radical entities in solution. Depending on the solvent, the prepared biradical crystallized in two different forms.

Pseudorotation in radical cations of low-symmetric decalin molecules.

Adiabatic potential energy surfaces (PES) of isomeric decalin cations have been found to be the pseudorotational surfaces due to avoided crossing that is typical for the highly symmetric Jahn-Teller active ions rather than for low-symmetric bicyclic systems. According to the UB3LYP/6-31G* data, the height of the barrier to pseudorotation is less than 2 kcal/mol for the trans isomer and about 9 kcal/mol for the cis isomer. Another peculiarity of the cis isomer PES is that the structure of minimum energy lies beyond the pseudorotation gutter.
