Article Synopsis
  • The SERENA-2 trial investigates the efficacy of camizestrant, a new oral selective estrogen receptor degrader, compared to the traditional injectable SERD, fulvestrant, in treating advanced hormone receptor-positive breast cancer in post-menopausal women.
  • This phase 2 trial includes patients who have experienced disease progression after previous endocrine therapies and assesses different dosages of camizestrant against fulvestrant, focusing on progression-free survival rates as the primary outcome.
  • Conducted across 74 centers worldwide, the study also monitors the safety and side effects of the treatments among all participants who received at least one dose.

Article Synopsis
  • The study explores how low levels of HER2 expression affect breast cancer treatment outcomes and the success of hormone therapy in patients.
  • It analyzes data from a large international study called PALLAS, which included over 5,000 patients to see how HER2 status impacts survival rates and response to the drug palbociclib.
  • The research found that HER2-low levels are common and vary between countries, but HER2 status didn’t significantly change the success rate of hormone therapy in the patients.

Background: Currently, it remains unknown whether there is an association between body mass index (BMI) and complications during screening colonoscopy; hence, it remains unclear whether BMI should be considered a risk factor in pre-procedural assessments. The aim of this study was to compare mortality and unplanned hospitalization rates before and after colonoscopy stratified by patients' BMI.

Material And Methods: This was a retrospective cohort study of individuals who underwent screening colonoscopy as part of the Polish Colonoscopy Screening Program (PCSP).

Despite new treatment strategies, graft-versus-host disease (GvHD) remains a formidable complication after allogeneic hematopoietic stem cell transplantation (HSCT). This study aimed to investigate the impact of polymorphisms and expression of MICA and NKG2D receptor on the development of GvHD in allogeneic HSCT recipients. Soluble MICA (sMICA) concentration was measured in serum collected 30 days after transplantation and the genetic variability of MICA and NKG2D genes was evaluated.

Article Synopsis
  • - Capivasertib, a selective pan-AKT inhibitor, was shown to significantly improve progression-free survival when added to fulvestrant compared to fulvestrant alone in patients with advanced breast cancer (P < 0.001), specifically those who had previously experienced disease progression on aromatase inhibitors.
  • - In a randomized trial with 708 patients, individuals received either capivasertib plus fulvestrant or a placebo plus fulvestrant, with safety analyses revealing common adverse events (AEs) like diarrhea, rash, and hyperglycemia associated with capivasertib treatment.
  • - Among 705 patients analyzed, 72.4% experienced diarrhea, while 38% had a rash

Background: T-cell immunoglobulin and mucin-domain containing-3 (TIM-3) molecule is a key regulator of the immune response by exerting an inhibitory effect on various types of immune cells. Understanding the role of TIM-3 in hematopoietic stem cell transplantation (HSCT) may improve transplant outcomes. Our study evaluated the potential association between TIM-3 polymorphisms, namely rs1036199 (A > C) or rs10515746 (C > A), changes which are located in exon 3 and the promoter region of the TIM-3 gene, and post-HSCT outcomes.

Article Synopsis
  • European health systems can improve health by using more technology to prevent diseases and spot them early.
  • Right now, they're not using these tools effectively, which means patients aren't getting the best diagnosis or access to new treatments.
  • The study looks into how different countries in Europe are handling new health technologies and genetics, and suggests it's a good time to rethink how to use these tools better in healthcare.

The aim of the present study was to determine the associations between the genetic variability and the expression and the risk of development of post-transplant complications after allogeneic hematopoietic stem cell transplantation (HSCT). HSCT recipients and their donors were genotyped for two polymorphisms (rs1065075, rs3828903). Moreover, the expression of a soluble form of MICB was determined in the recipients' serum samples after transplantation using the Luminex assay.

Introduction Patient-reported outcome measurements (PROMs) are gaining considerable popularity as tools to assess the effectiveness of the treatment in plastic surgery, being a complement to surgical outcomes. The SCAR-Q questionnaire has been recently developed for patients with surgical, traumatic, and burn scars. Aim The study aims to describe the process of translation and linguistic validation of the scar questionnaire (SCAR-Q) for use in Polish patients undergoing scar treatment.

Transplantation of hematopoietic stem cells (HSCT) is a procedure commonly used in treatment of various haematological disorders which is associated with significantly improved survival rates. However, one of its drawbacks is the possibility of development of post-transplant complications, including acute and chronic graft-versus-host disease (GvHD) or CMV infection. Various studies suggested that NK cells and their receptors may affect the transplant outcome.

Background: Evidence for the efficacy of combined PD-1 and HER2 blockade with chemotherapy on progression-free and overall survival in HER2-positive gastro-oesophageal cancer is scarce. The first interim analysis of the randomised, phase 3 KEYNOTE-811 study showed a superior objective response with pembrolizumab compared with placebo when added to trastuzumab plus fluoropyrimidine and platinum-based chemotherapy. Here, we report results from protocol-specified subsequent interim analyses of KEYNOTE-811.

  • Tenosynovial giant cell tumour (TGCT) is a rare tumor that grows around joints and tendons, with two types: nodular (less aggressive) and diffuse (more aggressive).
  • TGCT can lead to chronic pain and negatively affect a person's quality of life, though it's not usually life-threatening.
  • A global meeting in June 2022 brought together experts and patients to establish better treatment guidelines for TGCT, given the challenges in making consistent treatment choices worldwide.

Background: Although colonoscopy is widely used as a screening test to detect colorectal cancer, its effect on the risks of colorectal cancer and related death is unclear.

Methods: We performed a pragmatic, randomized trial involving presumptively healthy men and women 55 to 64 years of age drawn from population registries in Poland, Norway, Sweden, and the Netherlands between 2009 and 2014. The participants were randomly assigned in a 1:2 ratio either to receive an invitation to undergo a single screening colonoscopy (the invited group) or to receive no invitation or screening (the usual-care group).

Background: Current guidelines provide weak recommendations to treat small (<2 cm) non-functional pancreatic neuroendocrine tumors with low Ki-67 proliferation index either by resection or clinical follow-up. However, there is a lack of consensus regarding the minimal size of pNET, which allows EUS-guided biopsy with high enough diagnostic accuracy for stratification.

Methods: We conducted a retrospective, bicentric analysis of patients who had undergone EUS-guided pNET sampling in two tertiary care Endoscopy Units in Germany and Poland.
