Background: Occult papillary thyroid carcinoma (PTC) refers to the PTC accidentally found due to its occult lesions. Classic, as the most common subtype of PTC, is usually considered to have a low degree of malignancy and a favorable prognosis. Currently, the American Thyroid Association Management Guidelines adopted active surveillance (AS) as an alternative to immediate surgery in some low-risk PTC patients with less than 1 cm in diameter.

A perspective on the application of macrocyclic design strategies in antitumor drugs.

The macrocyclization of inhibitors has become gradually favored as a new approach for drug design in the field of anticancer agents, since the recent approvals of lorlatinib, pacritinib, and repotrectinib, and feasibility of macrocyclic modification to improve inhibitor drug-like properties has also been confirmed. Macrocycles are receiving increasing attention due to their enhanced binding affinity, target selectivity, and pharmacokinetic properties through conformational constraints. Therefore, this review summarizes various strategies for improving drug-like properties in macrocyclization and structural optimization, and reveals that macrocyclization is a new favorable strategy for drug design, aiming to provide insights for the drug discovery in different targets.

Assessment of textbook oncologic outcome after neoadjuvant chemotherapy and interval debulking surgery for advanced ovarian cancer.

Aim: To examine the prognostic impact of textbook oncologic outcome (TOO) in patients with advanced ovarian cancer undergoing primary chemotherapy, along with identifying the risk factors for TOO failure.

Methods: Patients who underwent neoadjuvant chemotherapy followed by interval debulking surgery for advanced ovarian cancer at a tertiary center between 2014 and 2019 were retrospectively reviewed. TOO was defined as complete cytoreduction, no severe complications, no prolonged hospital stay, no readmission, no delayed initiation of adjuvant chemotherapy, and no 90-day mortality.

This study aimed to investigate the relationship between the TyG index and both all-cause and CVD mortality in this population within the United States. Our study recruited 1778 adults with diabetes aged < 65 years without cardiovascular disease from the National Health and Nutrition Examination Survey (NHANES) 2003-2018.

This research highlights the critical demand for advanced prognostic tools by creating a model centered on aging-related genes (ARGs) to improve prediction and treatment strategies for lung adenocarcinoma (LUAD). By leveraging datasets from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO), we developed a prognostic model that integrates 14 ARGs using the least absolute shrinkage and selection operator (LASSO) alongside Cox regression analyses.

Group 2 innate lymphoid cells (ILC2s) produce the type 2 cytokines IL-5 and IL-13 and contribute to type 2 immune responses, such as allergic airway inflammation. However, specific drugs, especially traditional Chinese medicines, that target lung ILC2s have rarely been reported. Here, we demonstrate that triptolide ameliorates allergic airway inflammation by suppressing ILC2 activation.
