Chromosomal rearrangements and recurrent gene fusions were previously presumed to be the primary oncogenic mechanisms of hematological malignancies. However, the discovery of gene fusions in different cancers has opened new horizons to comprehensively investigate how cell type-specific fusion oncoproteins modulate signaling cascades. Prostate cancer (PCa) is a multifaceted and therapeutically challenging disease, and functional genomics have helped us develop a better understanding of the mechanisms underlying prostate carcinogenesis, castration-resistant PCa, and metastasis.

Recent studies strongly suggest that gut microbiome can influence brain functions and contribute to the development of Alzheimer's disease (AD). However, reported changes in the gut microbiomes in AD patients from different countries are not similar, and more research is needed to reveal the relationships between human microbiomes and AD in diverse ethnic populations. There is also an assumption that microbiome-associated peripheral inflammation might drive the development of sporadic AD.

Article Synopsis
  • The study investigates how genetic variations in certain genes affect the metabolism and effects of the blood thinner dabigatran in patients with atrial fibrillation.
  • Using TaqMan technology, the research analyzed genotypes from 150 patients and found significant differences in dabigatran concentrations based on specific gene polymorphisms.
  • The findings suggest that polymorphisms rs8192935 and rs2244613 influence dabigatran's pharmacodynamics and pharmacokinetics, particularly in the Kazakh population.

Oral Microbiome Stamp in Alzheimer's Disease.

Pathogens

February 2024

Center for Life Sciences, National Laboratory Astana, Nazarbayev University, Astana Z05H0P9, Kazakhstan.

Recent studies have suggested that periodontal disease and alterations in the oral microbiome may be associated with cognitive decline and Alzheimer's disease (AD) development. Here, we report a case-control study of oral microbiota diversity in AD patients compared to healthy seniors from Central Asia. We have characterized the bacterial taxonomic composition of the oral microbiome from AD patients (n = 64) compared to the healthy group (n = 71) using 16S ribosomal RNA sequencing.

Background: Type 2 diabetes mellitus (T2DM) is associated with sleep disorders, which in turn may lead to anxiety and depression. Sleep deprivation impairs glucose metabolism causing a decrease in insulin sensitivity and glucose tolerance. The present study aimed to determine the association of insomnia with anxiety and depression in patients with T2DM.

Nowadays, direct oral anticoagulants (DOACs) are the first-line anticoagulant strategy in patients with non-valvular atrial fibrillation (NVAF). We aimed to identify the influence of polymorphisms of the genes encoding P-glycoprotein () and carboxylesterase 1 () on the variability of plasma concentrations of DOACs in Kazakhstani patients with NVAF. We analyzed polymorphisms rs4148738, rs1045642, rs2032582 and rs1128503 in and rs8192935, rs2244613 and rs71647871 genes and measured the plasma concentrations of dabigatran/apixaban and biochemical parameters in 150 Kazakhstani NVAF patients.

Acquired Hallux Varus. Etiology, Pathology, Classification, and Treatment Algorithm. Literature Review.

Arch Razi Inst

December 2022

Department of Traumatology and Orthopedics, NJSC Medical University Astana, Beibitshilik St 49/A, Astana 010000, Kazakhstan.

Iatrogenic hallux varus is formed due to an imbalance between the bone, tendon, and ligamentous-articular structures in the first metatarsophalangeal joint (MJ), with the progression of the medial deviation of the big toe. A secondary factor is an imbalance between excessive medial tension and weakness or excessive soft tissue release of lateral capsular-ligamentous structures. The article is devoted to a rare but no less complex postoperative complication of hallux valgus deformity, acquired hallux varus.

Study of gut microbiota alterations in Alzheimer's dementia patients from Kazakhstan.

Sci Rep

September 2022

Laboratory of Bioengineering and Regenerative Medicine, Center for Life Sciences, National Laboratory Astana, Nazarbayev University, Nur-Sultan, Kazakhstan.

We have investigated the diversity and composition of gut microbiotas isolated from AD (Alzheimer's disease) patients (n = 41) and healthy seniors (n = 43) from Nur-Sultan city (Kazakhstan). The composition of the gut microbiota was characterized by 16S ribosomal RNA sequencing. Our results demonstrated significant differences in bacterial abundance at phylum, class, order, and genus levels in AD patients compared to healthy aged individuals.

Background: In patients with Hirschsprung's disease (HD), persistent obstructive symptoms may develop after surgery. The causes of mechanical obstruction may be a retraction of the pulled-through bowel due to insufficient mobilization of the mesentery or impaired blood supply in the area of the coloanal anastomosis in the case of excessive ligation of the supplying vessels. Leakage and stenosis are formidable complications and require repeated surgical intervention.

  • The study investigates how factors like GDP, prevalence of overweight/obesity, NCD mortality, and vaccination rates impact COVID-19 morbidity and mortality across various countries.
  • Data analysis from 218 countries reveals significant correlations between COVID-19 rates and various social and economic indicators, such as GDP and obesity statistics.
  • Increased vaccination rates have a positive effect on reducing new COVID-19 cases, although the statistical significance is marginal, especially in non-island countries compared to global averages.

The prevalence of sleep disorders in the general population is quite high. At the same time, changes in sleep duration, sleep apnoea syndrome, fragmentation and other changes in sleep are significant factor of risk for the development of diabetes mellitus type 2, and also have a negative impact on the ability to achieve satisfactory compensation in these patients. The article presents up-to-date information on the association between sleep disorders and diabetes mellitus type 2.

June 2018

JSC "Medical University Astana", Department of Children's Diseases №1, Kazakhstan.

In connection with the urgency of early diagnosis of severe combined immune deficiency, the case from the clinical practice of the National Science Center for Maternity and Childhood, Astana, was considered. This clinical case indicates that it is difficult to verify the diagnosis of severe combined failure, as the child was hospitalized in city clinics with various infectious diseases before receiving the right treatment more than 10 times in 4 months of life. Also, this clinical example confirms that the most effective method of treating this disorder at the present stage is bone marrow transplantation.
