Long-chain fatty acid (LCFA) oxidation has been shown to play an important role in interleukin-4 (IL-4)-mediated macrophage polarization (M(IL-4)). However, many of these conclusions are based on the inhibition of carnitine palmitoyltransferase-1 with high concentrations of etomoxir that far exceed what is required to inhibit enzyme activity (EC < 3 μM). We employ genetic and pharmacologic models to demonstrate that LCFA oxidation is largely dispensable for IL-4-driven polarization.

We report on a patient with deep brain stimulation (DBS) of the subthalamic nucleus (STN) for Parkinson's disease (PD) who developed pathologic crying (PC) immediately postoperatively. A spread of DBS current to adjacent cortico-ponto-cerebellar pathways might be responsible. The phase III, randomized, open-label CheckMate 025 study showed an overall survival benefit in patients with advanced renal cell carcinoma treated with nivolumab versus everolimus. Here, we review the health-related quality of life (HRQoL) results of this trial, in which nivolumab was associated with significant improvements in HRQoL, with more patients having a clinically meaningful HRQoL improvement and a shorter time to onset of improvement compared with everolimus. Further exploratory analysis suggests a positive correlation between baseline HRQoL scores and overall survival.

Although it is known that MyD88-deficient mice are highly susceptible to a wide range of bacterial infections, the cell type-specific contribution of MyD88 in protecting the host against intestinal bacterial infection is only poorly understood. Hepatitis C virus (HCV) displays a restricted host species tropism and only humans and chimpanzees are susceptible to infection. A robust immunocompetent animal model is still lacking, hampering mechanistic analysis of virus pathogenesis, immune control, and prophylactic vaccine development. The closest homolog of HCV is the equine nonprimate hepacivirus (NPHV), which shares similar features with HCV and thus represents an animal model to study hepacivirus infections in their natural hosts.

Even though the virus is mainly transmitted via mosquitos, human-to-human transmission has been described. Infectious viral particles have been detected in breast milk of infected women which raised concerns regarding the safety of breastfeeding in areas of Zika virus transmission or in case of a suspected or confirmed Zika virus infection.
