Background: Neuropathic adverse events occur frequently in linezolid-containing regimens, some of which remain irreversible after drug discontinuation.

Objective: We aimed to identify and validate a host RNA-based biomarker that can predict linezolid-associated neuropathy before multidrug-resistant/rifampicin-resistant tuberculosis (MDR/RR-TB) treatment initiation and to identify genes and pathways that are associated with linezolid-associated neuropathy.

Methods: Adult patients initiating MDR/RR-TB treatment including linezolid were prospectively enrolled in 3 independent cohorts in Germany.

Article Synopsis
  • In 2020, nearly 500,000 people were diagnosed with rifampicin-resistant tuberculosis (RR-TB), leading to an estimated 6.9 million disability-adjusted life years (DALYs) lost, impacting mostly TB survivors.
  • The burden of RR-TB was notably higher in former Soviet Union and southern African countries, with an average loss of 17 DALYs per RR-TB patient—34% more than for regular TB cases.
  • The findings highlight the urgent need for enhanced post-treatment care and more funding to prevent the spread of RR-TB, due to its significant long-term health effects on survivors.

Despite the advent of new diagnostics, drugs and regimens, tuberculosis (TB) remains a global public health threat. A significant challenge for TB control efforts has been the monitoring of TB therapy and determination of TB treatment success. Current recommendations for TB treatment monitoring rely on sputum and culture conversion, which have low sensitivity and long turnaround times, present biohazard risk, and are prone to contamination, undermining their usefulness as clinical treatment monitoring tools and for drug development.

The accumulated evidence maps the COVID-19 pandemic's diverse impacts on sexual and reproductive health (SRH); however, the precise changes in sexual behaviours and the underlying causes producing these changes are rarely considered. This study is aimed at assessing the changes in sexual behaviours during the COVID-19 pandemic in Germany, using quantitative methods, and it is also aimed at identifying the underlying reasons, using qualitative methods. It is a part of the broader I-SHARE project, which administered a cross-sectional online survey in 33 countries to describe the effects of the COVID-19 restrictions on different aspects of SRH.

An estimated 58 million people have survived tuberculosis since 2000, yet many of them will suffer from post-tuberculosis lung disease (PTLD). PTLD results from a complex interplay between organism, host, and environmental factors and affects long-term respiratory health. PTLD is an overlapping spectrum of disorders that affects large and small airways (bronchiectasis and obstructive lung disease), lung parenchyma, pulmonary vasculature, and pleura and may be complicated by co-infection and haemoptysis.
