Ruthenium(ii) complexes are attracting significant research attention as a promising class of photosensitizers (PSs) in photodynamic therapy (PDT). Having previously reported the synthesis of two novel Ru(ii)-polypyridyl-1,8-naphthalimide Tröger's base compounds 1 and 2 with interesting photophysical properties, where the emission from either the Ru(ii) polypyridyl centres or the naphthalimide moieties could be used to monitor binding to nucleic acids, we sought to use these compounds to investigate further and in more detail their biological profiling, which included unravelling their mechanism of cellular uptake, cellular trafficking and cellular responses to photoexcitation. Here we demonstrate that these compounds undergo rapid time dependent uptake in HeLa cells that involved energy dependent, caveolae and lipid raft-dependent mediated endocytosis, as demonstrated by confocal imaging, and transmission and scanning electron microscopy.

Waterlogging leads to major crop losses globally, particularly for waterlogging-sensitive crops such as barley. Waterlogging reduces oxygen availability and results in additional stresses, leading to the activation of hypoxia and stress response pathways that promote plant survival. Seasonal precipitation forecasting is highly challenging for the northwest fringes of Europe due to complex dynamical drivers. The dense glycan shield is an essential feature of the SARS-CoV-2 spike (S) architecture, key to immune evasion and to the activation of the prefusion conformation. Recent studies indicate that the occupancy and structures of the SARS-CoV-2 S glycans depend not only on the nature of the host cell, but also on the structural stability of the trimer; a point that raises important questions about the relative competence of different glycoforms. Moreover, the functional role of the glycan shield in the SARS-CoV-2 pathogenesis suggests that the evolution of the sites of glycosylation is potentially intertwined with the evolution of the protein sequence to affect optimal activity.

A 250-year (1766-2016) archive of reconstructed river flows is presented for 51 catchments across Ireland. By leveraging meteorological data rescue efforts with gridded precipitation and temperature reconstructions, we develop monthly river flow reconstructions using the GR2M hydrological model and an Artificial Neural Network. Uncertainties in reconstructed flows associated with hydrological model structure and parameters are quantified.

Time series homogenization for 299 of the available precipitation records for the island of Ireland (IENet) was performed. Four modern relative homogenization methods, that is, HOMER, ACMANT, CLIMATOL and AHOPS were applied to this network of station series where contiguous intact monthly records range from 30 to 70 years within the base period 1941-2010. Break detection results are compared between homogenization methods, and coincidences with available documentary information (metadata) were analysed.

This study develops a coherent framework to detect those catchment types associated with a high risk of maladaptation to future flood risk. Using the "scenario-neutral" approach to impact assessment the sensitivity of Irish catchments to fluvial flooding is examined in the context of national climate change allowances. A predefined sensitivity domain is used to quantify flood responses to +2 °C mean annual temperature with incremental changes in the seasonality and mean of the annual precipitation cycle.

Non-ribosomal peptide (NRP) synthesis in fungi requires a ready supply of proteogenic and non-proteogenic amino acids which are subsequently incorporated into the nascent NRP via a thiotemplate mechanism catalyzed by NRP synthetases. Substrate amino acids can be modified prior to or during incorporation into the NRP, or following incorporation into an early stage amino acid-containing biosynthetic intermediate. These post-incorporation modifications involve a range of additional enzymatic activities including but not exclusively, monooxygenases, methyltransferases, epimerases, oxidoreductases, and glutathione S-transferases which are essential to effect biosynthesis of the final NRP.
