To determine the disease spectrum and genetic characteristics of inborn errors of metabolism (IEM) in Xinjiang province in the northwest of China, 41,690 newborn babies were screening by tandem mass spectrometry from November 2018 to December 2021. Of these, 57 newborn babies were referred for genetic analysis by next-generation sequencing, which was validated by Sanger sequencing. A total of 36 newborn babies and one relative were diagnosed with IEM, and the overall positive predictive value was 29.

Upregulation of PD-L1 contributes to improving the apoptosis of granulosa cells via the PI3K/AKT pathway in PCOS.

J Assist Reprod Genet

December 2024

State Key Laboratory of Pathogenesis, Prevention and Treatment of High Incidence Diseases in Central Asia, Xinjiang Medical University, Urumqi, China.

Article Synopsis
  • Polycystic ovary syndrome (PCOS) is a major cause of infertility linked to the apoptosis (cell death) of ovarian granulosa cells, and the role of the PD-1/PD-L1 pathway in this condition is not well understood.
  • Researchers analyzed RNA sequencing data from PCOS patients and established a mouse model to study the effects of PD-L1 intervention on ovarian granulosa cell apoptosis, using various molecular biology techniques.
  • The study found that the PD-L1 pathway can activate the PI3K/AKT signaling, reducing cell death in granulosa cells, suggesting a new potential strategy for treating apoptosis in PCOS.

Adipose-derived stromal cells (ADSCs) are commonly used in regenerative medicine, but the genetic features of their development into neuronal cells are unknown. This study used single-cell RNA sequencing (scRNA-seq) to reveal gene expression changes during ADSCs to neuronal differentiation. Sequencing of the ADSCs group, the prei-1d group, and the induction 1 h, 3 h, 5 h, 6 h, and 8 h groups was performed using the BD Rhapsody platform.

Fermentation engineering is critical for mass-producing chemicals, food additives, and medicines, where optimal culture conditions maximize microbial growth and metabolite production. Although monitoring bacterial growth during fermentation is critical, there is a lack of a non-invasive and sensitive method to directly monitor the bacterial metabolism. In this paper, a novel optical monitoring method is proposed based on tunable diode laser absorption spectroscopy.

Background: Wilms tumor (WT) is the most common pediatric kidney cancer, with survival rates exceeding 90% in localized cases. However, advanced or recurrent WT remains difficult to treat due to poor prognosis and limited knowledge of its molecular mechanisms. Gene expression profiling has shown promise in identifying prognostic markers and therapeutic targets.

Background: Metabolic-associated steatotic liver disease (MASLD) is a novel term proposed in 2023 to replace non-alcoholic fatty liver disease (NAFLD) with the aim of better reflecting its pathogenesis and clinical manifestations. Vibration-controlled transient elastography (VCTE) is an evidence-based, non-invasive imaging device used to evaluate liver fat deposition and fibrosis. It can effectively detect liver fat infiltration greater than 5%, which is much higher than the previous ultrasound detection rate (it is difficult to detect liver fat deposition below 30%).
