Background: Larsucosterol is a DNA methyltransferase inhibitor in development for alcohol-associated hepatitis (AH), a disease for which there is no approved therapy.
Methods: In this phase 2b trial, patients with severe AH were randomly assigned 1:1:1 to receive 30 mg or 90 mg of larsucosterol or placebo; a second dose was administered after 72 hours if the patient remained hospitalized. All patients received supportive care as determined by investigators.

This scoping review explores the concepts of integrated healthcare, interprofessional collaboration, and healthcare funding within the context of primary and allied healthcare. A systematic database, internet, and manual search of included article reference lists sought published and gray literature. From an initial 8,122 papers, a total of 63 met the inclusion criteria and were assessed using a three-stage narrative synthesis that sought to meaningfully account for the complexity and heterogeneity of the included papers: (1) Preliminary analysis involved data extraction and mapping of key themes, including article, integration, collaboration, and funding characteristics; (2) Robustness evaluation involved critically appraising the methodological quality of the literature using the Crowe Critical Appraisal Tool, and the Johns Hopkins Nursing Evidence-based Practice Research Evidence Appraisal Tool, and Non-Research Evidence Appraisal Tool; and (3) Relationship exploration found that most primary and allied healthcare services still operate under fee-for-service funding arrangements that discourage the delivery of integrated collaborative, coordinated, and complex care, instead encouraging traditional siloed and hierarchical approaches that are linked to workload, remuneration, and job satisfaction inequalities between primary and allied healthcare professions.

Despite a high burden of osteoporosis and minimal trauma fractures worldwide, there is still a treatment gap in timely diagnosis and optimal treatment. There is also a lack of international consensus and guidelines on the management of bone fragility in premenopausal women. This review article provides an overview of the current understanding of factors impacting women's bone health across the adult lifespan, as well as dilemmas in the diagnosis, assessment and management of osteoporosis in premenopausal and postmenopausal women, premature ovarian insufficiency and bone health following breast cancer.

Adenosine-to-inosine (A-to-I) editing, is a highly prevalent posttranscriptional modification of RNA, mediated by the adenosine deaminases acting on RNA (ADAR) proteins. Mammalian transcriptomes contain tens of thousands to millions of A-to-I editing events. Mutations in ADAR can result in rare autoinflammatory disorders such as Aicardi-Goutières syndrome (AGS) through to irreversible conditions such as motor neuron disease, amyotrophic lateral sclerosis (ALS).

Approximately half of critically ill adults experience intensive care unit acquired weakness (ICUAW). Patients who develop ICUAW may have negative outcomes, including longer duration of mechanical ventilation, greater length of stay, and worse mobility, physical functioning, quality of life, and mortality. Early physical rehabilitation interventions have potential for improving ICUAW; however, randomized trials show inconsistent findings on the efficacy of these interventions.

Persistent post-surgical pain (PPSP) occurs in a proportion of patients following surgical interventions. Research suggests that specific microbiome components are important for brain development and function, with recent studies demonstrating that chronic pain results in changes to the microbiome. Consumption of a high fat, high sugar (HFHS) diet can drastically alter composition of the microbiome and is a modifiable risk factor for many neuroinflammatory conditions.

Influenza A virus (IAV) and influenza B virus (IBV) with similar symptoms of infection caused a serious disease burden and economic losses in annual epidemic season, so it is important to quickly and accurately detect and distinguish between IAV and IBV during influenza season. Herein, the quantum dot microspheres (QDMS) were synthesized and applied to lateral flow immunoassays (LFIA), and a point-of-care (POC) biosensor that can discriminately and simultaneously diagnose IAV and IBV within 10 min was established. A double-sandwich QDMS nanotags was synthesized by immobilizing hydrophobic quantum dots (QDs) with chemical bonding method on a silica sphere template with an outer silica shell protection showed excellent stability and high fluorescence.

The mechanisms linking maternal asthma (MA) exposure in utero and subsequent risk of asthma in childhood are not fully understood. Pathological airway remodelling, including reticular basement membrane thickening, has been reported in infants and children who go on to develop asthma later in childhood. This suggests altered airway development before birth as a mechanism underlying increased risk of asthma in children exposed in utero to MA.

Adolescence is a vulnerable period for the onset of mental disorders and risk behaviours. Based on the Health-Promoting Schools Framework, whole-school interventions offer a promising strategy in this developmentally-sensitive cohort, through championing a systems-based approach to promotion and prevention that involves the key stakeholders in an adolescent's life. The evidence-base surrounding the effectiveness of whole-school interventions, however, remains inconclusive, partly due to the insufficient number of studies in previous meta-analyses.
