Mental illness, affecting one in eight people worldwide, is often exacerbated by stigma-which can result in self-stigmatization, isolation, and loneliness and negatively impact access to health care, education, and social connection. Previous research has found that stigma is best reduced through a combination of education about the stigmatized population and intentional contact with individuals from that population. Observational studies play an increasingly important role in estimating causal effects of a treatment or an exposure, especially with the growing availability of routinely collected real-world data. To facilitate drawing causal inference from observational data, we introduce a conceptual framework centered around "four targets"-target estimand, target population, target trial, and target validity. We illustrate the utility of our proposed "four targets" framework with the example of buprenorphine dosing for treating opioid use disorder, explaining the rationale and process for employing the framework to guide causal thinking from observational data.

Transgender youth are disproportionately affected by HIV, particularly minoritized youth in the US south. To understand HIV service use among transgender youth, we interviewed 25 young racial and ethnic minority clients of four southern community-based HIV service organizations (CBOs), and CBO staff ( = 12), about service access and use. Angiotensin-I converting enzyme (ACE) regulates the levels of disparate bioactive peptides, notably converting angiotensin-I to angiotensin-II and degrading amyloid beta. We introduce Hydrogen-Exchange Experimental Structure Prediction (HX-ESP), a method that integrates hydrogen exchange (HX) data with molecular dynamics (MD) simulations to accurately predict ligand binding modes, even for targets requiring significant conformational changes. Benchmarking HX-ESP by fitting two ligands to PAK1 and four ligands to MAP4K1 (HPK1), and comparing the results to X-ray crystallography structures, demonstrated that HX-ESP successfully identified binding modes across a range of affinities significantly outperforming flexible docking for ligands necessitating large conformational adjustments. By objectively guiding simulations with experimental HX data, HX-ESP overcomes the long timescales required for binding predictions using traditional MD.

Osteopetrosis is a rare genetic disorder that leads to increased bone density and fragility due to dysfunctional osteoclasts, which can result in narrowed bone marrow spaces, hardened cartilage, and brittle bones. This condition frequently coexists with osteoarthritis among affected individuals. In 2022, the European Union put forward the REPowerEU plan in response to Russia's invasion of Ukraine, aiming at enhancing short-term energy security by diversifying imports and reducing natural gas demand while accelerating the deployment of renewable alternatives in the long term. Here, we quantify the life cycle environmental impacts of both REPowerEU's short-term measures, including the controversial extended coal-fired power plant operations, and how the first year of the crisis was managed in practice. We find that the policy measures' impact on greenhouse gas (GHG) emissions would be negligible, although they could have detrimental effects on other environmental categories.

Methods: Disease-free cancer survivors (>3 months since therapy and age > 18 years) treated with intensity modulated RT for early (T1-2, N0-2b) tonsillar cancer with pre- and post-therapy contrast-enhanced CT scans available were included. A mirror-image experiment: Sorting carbon nanotubes by L-DNA.

Here, we report a mirror-image experiment using naturally occurring D-DNA and its enantiomer L-DNA to sort a chiral mixture of single-wall carbon nanotubes (SWCNTs). We find that parity conservation leads to a robust experimental outcome: changing DNA chirality results in handedness inversion of the purified nanotube.

Background: Current methods of measuring disease progression of neurodegenerative disorders, including Parkinson's disease (PD), largely rely on composite clinical rating scales, which are prone to subjective biases and lack the sensitivity to detect progression signals in a timely manner. Digital health technology (DHT)-derived measures offer potential solutions to provide objective, precise, and sensitive measures that address these limitations. However, the complexity of DHT datasets and the potential to derive numerous digital features that were not previously possible to measure pose challenges, including in selection of the most important digital features and construction of composite digital biomarkers.
