Imprinting abnormalities pose a significant challenge in applications involving embryonic stem cells, induced pluripotent stem cells, and animal cloning, with no universal correction method owing to their complexity and stochastic nature. In this study, we targeted these defects at their source-embryos from same-sex parents-aiming to establish a stable, maintainable imprinting pattern de novo in mammalian cells. Using bi-paternal mouse embryos, which exhibit severe imprinting defects and are typically non-viable, we introduced frameshift mutations, gene deletions, and regulatory edits at 20 key imprinted loci, ultimately achieving the development of fully adult animals, albeit with a relatively low survival rate.

Dysregulation of genes encoding the homologous to E6AP C-terminus (HECT) E3 ubiquitin ligases has been linked to cancer and structural birth defects. One member of this family, the HECT-domain-containing protein 1 (HECTD1), mediates developmental pathways, including cell signaling, gene expression, and embryogenesis. Through GeneMatcher, we identified 14 unrelated individuals with 15 different variants in HECTD1 (10 missense, 3 frameshift, 1 nonsense, and 1 splicing variant) with neurodevelopmental disorders (NDDs), including autism, attention-deficit/hyperactivity disorder, and epilepsy.

The mechanics of this elongation, which requires the presomitic mesoderm (PSM), remain poorly understood. Purpose: To examine the relationship between dyadic coping and dyadic adjustment in patients with cervical cancer and their spouses in northwest China, and explore the mediating role of psychological resilience in this relationship.

Methods: A cross-sectional study was conducted from June to October 2024 in two tertiary hospitals in Xinjiang Province, China; 260 patients with cervical cancer and their spouses were enrolled. Participants independently completed demographic and clinical questionnaires as well as Chinese versions of the Resilience Scale, Dyadic Coping Inventory, and Locke-Wallace Marital Adjustment Scale.

Traffic signals, while reducing conflicts within intersections, often lead to stop-and-go behaviors in approaching vehicles, negatively impacting traffic flow in terms of safety, efficiency, and fuel consumption. Aimed at minimizing the traffic oscillations caused by traffic signals through Connected and Autonomous Vehicles (CAVs) and meeting real-time operational needs, this paper proposes a Risk-Based Adaptive Cruise Control (RACC). RACC designs the constraints of approaching a signalized intersection as expected risks, enabling compliance with all constraints while being adaptable to basic road scenarios.

There has been a recent influx in the development of novel measures of structural forms of discrimination, including structural racism, xenophobia, sexism, heterosexism, and cisgenderism. These systems of power and oppression are inherently interdependent and mutually constitutive, yet a paucity of research has investigated their joint impacts; this gap is likely reflective of the limited guidance that exists regarding how to effectively combine multiple measures of structural discrimination to examine their joint impacts on population health and health inequities. In this commentary, we seek to redress this by describing conceptual and methodologic considerations for population health researchers interested in conducting quantitative structural intersectionality research - an intersectionality-informed research approach focused on examining how systems of power and oppression intersect to shape population health and health inequities.

We solve high-dimensional steady-state Fokker-Planck equations on the whole space by applying tensor neural networks. The tensor networks are a linear combination of tensor products of one-dimensional feedforward networks or a linear combination of several selected radial basis functions. The use of tensor feedforward networks allows us to efficiently exploit auto-differentiation (in physical variables) in major Python packages while using radial basis functions can fully avoid auto-differentiation, which is rather expensive in high dimensions.

Ischemia/reperfusion injury (I/RI) following myocardial infarction, a leading cause of global morbidity and mortality, is characterized by detrimental oxidative stress and inflammation. In response, we proposed an I/RI alleviation strategy using the intravenous injection of spherical selenium nanoparticles (SeNPs) synthesized by a template method. Single-cell sequencing revealed these proposed SeNPs exhibited exceptional antioxidant and anti-inflammatory properties, disrupting the STAT1-ROS cycle, therefore preserving mitochondrial respiration and inhibiting caspase-mediated cardiomyocyte apoptosis.

Point mutations were introduced into specific leucine (L) amino acids within the K domain of SHORT VEGETATIVE PHASE (SVP), and their effects on the SVP-AP1 interaction were assessed. Yeast two-hybrid experiments and β-galactosidase activity assays demonstrated that SVP maintained its capacity to interact with APETALA1 (AP1) despite point mutations at the 108th, 116th, 119th, and 127th leucine residues, where leucine was substituted with alanine (A). However, the mutation of the leucine residue at position 124 to alanine abolished the interaction between SVP and AP1 regardless of whether the mutation was singular or combined with others.

Due to a lack of anatomical studies utilizing female specimens, it is unclear how the nociceptive innervation of the mouse heart compares between sexes. To address this, flat-mount preparations of the left and right atria of male and female mice were immunohistochemically labeled for calcitonin gene-related peptide (CGRP, a common marker for nociceptive nerves), imaged, and digitally traced in high quality. The results show that 1) A network of CGRP-IR axons densely innervated the right and left atria.
