Sex is a key variable in the regulation of human physiology and pathology. Many diseases disproportionately affect one sex: autoimmune diseases, such as systemic lupus erythematosus, are more common in women but more severe in men, whereas the incidence of other disorders such as gouty arthritis and malignant cancers is higher in men. Besides the pathophysiology, sex may also influence the efficacy of therapeutics; participants in clinical trials are still predominately men, and the side effects of drugs are more common in women than in men.

Aims: Atrial fibrillation (AF) is common in heart failure (HF) and negatively impacts outcomes. The role of ablation-based rhythm control in patients with AF and HF with preserved (HFpEF) or mildly reduced ejection fraction (HFmrEF) is not known. The CABA-HFPEF-DZHK27 (CAtheter-Based Ablation of atrial fibrillation compared to conventional treatment in patients with Heart Failure with Preserved Ejection Fraction) trial will determine whether early catheter ablation for AF can prevent adverse cardiovascular outcomes in patients with HFpEF or HFmrEF.

Article Synopsis
  • The study investigates whether DNA methylation profiling can serve as a prognostic marker to predict regrowth potential in pituitary adenomas after surgery.
  • It involved analyzing 42 pituitary macroadenoma samples using Illumina's MethylationEPIC array, leading to the identification of distinct DNA methylation clusters among tumors.
  • Although the results showed no statistically significant differences in regrowth rates between clusters, there were indications that certain methylation patterns might correlate with higher regrowth tendencies.

Article Synopsis
  • * Procedural success was high (99.2%), with minimal complications (2.7% need for bail-out stenting and only one embolization), and after an average follow-up of 1.5 years, acceptable rates of clinically driven target lesion revascularization (CD-TLR) and clinical success were observed.
  • * Atherectomy combined with drug-coated balloons (DCB) in the popliteal artery resulted in significantly lower CD-TLR rates compared to non-debulking strategies, underscoring its safety and efficacy

The diagnosis of ependymoma has moved from a purely histopathological review with limited prognostic value to an integrated diagnosis, relying heavily on molecular information. However, as the integrated approach is still novel and some molecular ependymoma subtypes are quite rare, few studies have correlated integrated pathology and clinical outcome, often focusing on small series of single molecular types. We collected data from 2023 ependymomas as classified by DNA methylation profiling, consisting of 1736 previously published and 287 unpublished methylation profiles.

The NLRP3 inflammasome transforms a wide variety of infectious and non-infectious danger signals that activate pro-inflammatory caspases, which promote the secretion of IL-1β and IL-18, and pyroptosis, a pro-inflammatory form of cell necrosis. Most published evidence documents the presence and importance of the NLRP3 inflammasome in monocytes, macrophages, and neutrophils during host defense and sterile forms of inflammation. In contrast, in numerous unbiased data sets, NLRP3 inflammasome-related transcripts are absent in non-immune cells.
