Article Synopsis
  • A study investigated how metabolic syndrome (MetS) traits and liver fibrosis affect the risk of chronic kidney disease (CKD) and end-stage renal disease (ESRD) in individuals with metabolic dysfunction-associated steatotic liver disease (MASLD).
  • Data from over 234,000 participants were analyzed, revealing that a significant portion had MASLD and MetS; certain traits like hypertension and type 2 diabetes increased CKD risk, especially when combined with advanced liver fibrosis.
  • The findings suggest that the presence and number of MetS traits, along with liver fibrosis, significantly raise the risk of CKD and the likelihood of developing ESRD over time.

Revealing the structural microenvironment of high metastatic risk uveal melanomas following decellularisation.

Sci Rep

November 2024

Liverpool Ocular Oncology Research Group, Department of Eye and Vision Science, Institute of Life Course and Medical Science, University of Liverpool, 3rd Floor William Henry Duncan Building, West Derby Street, Liverpool, L7 8TX, UK.

Uveal melanoma (UM) is a rare aggressive intraocular tumour that spreads most commonly to the liver in tumours with loss of one copy of chromosome 3 (HR-M3); current treatments for metastatic disease remain largely ineffective. Pre-clinical research is increasingly using three-dimensional models that better recapitulate the tumour microenvironment (TME). One aspect of the TME is the acellular extracellular matrix (ECM) that influences cell proliferation, migration and response to therapy.

Glaucoma is a sight threatening neurodegenerative condition of the optic nerve head associated with ageing and marked by the loss of retinal ganglion cells. Mitochondrial dysfunction plays a crucial role in the pathogenesis of neurodegeneration in the most prevalent type of glaucoma: primary open angle glaucoma (POAG). All previous mitochondrial genome sequencing studies in POAG analyzed mitochondrial DNA (mtDNA) isolated from peripheral blood leukocytes and have not evaluated cells derived from ocular tissue, which better represent the glaucomatous disease context.

Article Synopsis
  • The study investigates the relationship between graft-versus-host disease (GVHD) and graft-versus-leukemia (GVL) effects in acute myeloid leukemia (AML) patients after stem-cell transplantation (SCT), specifically comparing standard and post-transplant cyclophosphamide (PTCy) methods.
  • Results show that while GVHD correlates with lower relapse rates and higher non-relapse mortality (NRM) and lower overall survival (OS) in standard cases, following PTCy, GVHD does not reduce relapse.
  • The findings indicate that although GVHD remains a significant factor in SCT outcomes, its role changes depending on the type of prophylaxis used, particularly diminishing its association with relapse in the PTC

Optimal dosing in pregnant and lactating women requires an understanding of the pharmacokinetics in the mother, fetus, and breastfed infant. Physiologically-based pharmacokinetic (PBPK) modeling can be used to simulate untested scenarios and hence supplement clinical data to support dosing decisions. A PBPK model for the antiretroviral dolutegravir (mainly metabolized by UGT1A1) was verified using reported exposures in non-pregnant healthy volunteers, pregnant women, and the umbilical cord, lactating mothers, and breastfed neonates.
