Let-7b acts as a tumor suppressor in osteosarcoma via targeting IGF1R.

Oncol Lett

February 2019

Department of Orthopedics, Linyi Central Hospital, Linyi, Shandong 276400, P.R. China.

Article Synopsis
  • MicroRNAs play essential roles in cancer development by regulating protein synthesis and mRNA stability, with Let-7b being of particular interest in osteosarcoma (OS).
  • The study found that Let-7b levels were lower in OS tissues and cell lines compared to normal tissues, and overexpressing Let-7b inhibited the growth and spread of OS cells by targeting the insulin-like growth factor 1 receptor (IGF1R).
  • The findings suggest that Let-7b acts as a suppressor in OS, reducing IGF1R levels and indicating a potential new therapeutic target for preventing metastasis in this type of cancer.

August 2019

State Key Laboratory of Pharmaceutical Biotechnology, School of Life Sciences, Nanjing University, No. 163 Xianlin Road, Nanjing 210023, China.

Glycyrrhizic acid (GA), a triterpene isolated from the roots and rhizomes of licorice, named Glycyrrhiza glabra, is the principal bioactive ingredient of anti-viral, anti-inflammatory and hepatoprotective effects. GA has been used in the clinical treatment of hepatitis, bronchitis, gastric ulcer, AIDS (acquired immunodeficiency syndrome), certain cancers and skin diseases. It has a direct effect on anti-HBV (hepatitis B virus) via affecting the HBsAg (hepatitis B surface antigen) to extracellular secretion, improving liver dysfunction in patients with chronic hepatitis B, and ultimately improving the immune status of HBV.

We attempted to explore the possible effects of SOCS3 (suppressor of cytokine signaling 3)-modified bone marrow mesenchymal stem cells (BMSCs) on the hypoxic injury of rat adrenal gland pheochromocytoma (PC-12) cells. PC12 cells were cultured with EGFP (enhanced green fluorescent protein)-BMSCs and SOCS3-BMSCs respectively under hypoxia in vitro and classified into control, hypoxia, EGFP-BMSCs, and SOCS3-BMSC groups. CCK-8, Hoechst 33258 staining, and Annexin V-FITC/PI staining were assessed to measure the viability and apoptosis of hypoxia-induced PC12 cells.

In this study, a novel strategy for amplified electrochemiluminescence (ECL) detection of DNA was proposed based on excellent ECL activity of graphene quantum dots (GQDs) coupled with multiple cycling amplification technique. A new type of graphene QDs with well ECL property and uniform size were firstly synthesized, then the graphene QDs were assembled on the electrode by poly (diallyldimethylammonium chloride) (PDDA)-graphene oxide (GO) nanocomposites, which could greatly improve ECL signal and stability of QDs. A novel signal-on ECL biosensor for DNA analysis was designed by using ECL quenching of gold nanoparticles (NPs) to graphene QDs combined with endonuclease-aided cyclic amplification strategy.

Ubiquitin specific peptidase 11 (USP11) is a deubiquitinating enzyme (DUB) and has been observed to promote mammary tumor initiation and progression. Several reports have shown that USP11 plays an important role in cancer development. Previous research has shown that USP11 is upregulated in breast cancer, but the function of USP11 in ovarian cancer has not been determined to date.
