CIC fusions have been described in two different central nervous system (CNS) tumor entities. On one hand, fusions of CIC or ATXN1 genes belonging to the same complex of transcriptional repressors, were reported in the CIC-rearranged, sarcoma (SARC-CIC). The diagnosis of this tumor type, which was recently added to the World Health Organization (WHO) Classification of CNS tumors, is difficult mainly because the data concerning its histopathology (as compared to its soft tissue counterpart), immunoprofile, and clinical as well as radiological characteristics are scarce in the literature.

Article Synopsis
  • Subcortical brain structures play a crucial role in various developmental and psychiatric disorders, and a study analyzed brain volumes in 74,898 individuals, identifying 254 genetic loci linked to these volumes, which accounted for up to 35% of variation.
  • The research included exploring gene expression in specific neural cell types, focusing on genes involved in intracellular signaling and processes related to brain aging.
  • The findings suggest that certain genetic variants not only influence brain volume but also have potential causal links to conditions like Parkinson’s disease and ADHD, highlighting the genetic basis for risks associated with neuropsychiatric disorders.

Brain PET Imaging in the Presurgical Evaluation of Drug-Resistant Focal Epilepsy.

PET Clin

January 2025

King's College London & Guy's and St Thomas' PET Centre, School of Biomedical Engineering and Imaging Sciences, King's College London, Office Suite 6, 4th Floor Lambeth Wing, London, UK; St Thomas' Hospital, Westminster Bridge Road, London SE1 7EH, UK.

Presurgical evaluation aims to localize the seizure onset zone (SOZ) for a tailored resection. Interictal [F]fluorodeoxyglucose PET is now an established test to lateralize and/or localize the SOZ, particularly if MR imaging is negative or if the noninvasive assessment shows discrepancies. PET can show hypometabolic areas associated with SOZ and the potential altered metabolic brain networks.

P21 activated kinase 6 (PAK6) is a serine-threonine kinase with physiological expression enriched in the brain and overexpressed in a number of human tumors. While the role of PAK6 in cancer cells has been extensively investigated, the physiological function of the kinase in the context of brain cells is poorly understood. Our previous work uncovered a link between PAK6 and the Parkinson's disease (PD)-associated kinase LRRK2, with PAK6 controlling LRRK2 activity and subcellular localization via phosphorylation of 14-3-3 proteins.

Migraine treatment: Position paper of the French Headache Society.

Rev Neurol (Paris)

December 2024

Department of Neurology, Gui-de-Chauliac Hospital, CHU de Montpellier, University of Montpellier, 34000 Montpellier, France.

The French migraine management recommendations were published in 2021. However, in the last three years, new data have come to light and new drugs have been approved (eptinezumab, rimegepant and atogepant) by the European Medicines Agency that require us to take a position on their use and to update certain elements of the recommendations. The first important message concerns the position of the French Headache Society on the use of preventive treatments (monoclonal antibodies and gepants) targeting the calcitonin gene-related peptide (CGRP) pathway.

Article Synopsis
  • - High density microelectrode arrays (HD-MEAs) can record activity from many neurons at once, but since they don't show the physical location of neurons, algorithms are needed to track their positions as they move away from the recording equipment.
  • - This study compared several algorithms for estimating neuron locations from MEA data, finding that the grid-based algorithm performed best overall, while the center-of-mass method was cheaper but less accurate, and monopolar methods had good precision but high variability and cost.
  • - The results emphasize the need for researchers to choose the right localization techniques for better neuron tracking in their studies, as previous methods had not been rigorously validated against known locations.

Gender differences in the observation of gesture direction: a physiological study.

Sci Rep

October 2024

Univ. Lille, CNRS, UMR 9193 - SCALab - Sciences Cognitives et Sciences Affectives, F-59000, Lille, France.

Intransitive gestures are expressive and symbolic, whereas pantomimes are object-related actions. These gestures convey different meanings depending on whether they are directed toward (TB) or away from the body (AB). TB gestures express mental states (intransitive) or hygiene/nutritional activities (pantomime), while AB gestures modify the behaviour of the observer (intransitive) or demonstrate tool use with an object (pantomime).

Plasma membrane calcium ATPases (PMCAs) encoded by ATP2B genes have been implicated in Mendelian diseases with ataxia, dystonia, and intellectual disability. Work to date has shown that ATP2B2 (encoding PMCA2) is required for synaptic function and Purkinje-cell integrity in the cerebellum. A recent case series has linked ATP2B2 to a novel entity, characterized by neurodevelopmental and movement phenotypes, in only seven individuals.
