Loss-of-Function Variants in CUL3 Cause a Syndromic Neurodevelopmental Disorder.

Ann Neurol

September 2024

Department of Medical Genetics, Center for Medical Genetics, School of Basic Medical Sciences, Peking University, Beijing, China.

Article Synopsis
  • - This study examines the link between rare variants in the cullin-3 ubiquitin ligase (CUL3) gene and neurodevelopmental disorders (NDDs), gathering data from multiple centers to explore genetic mutations and their clinical impacts.
  • - Researchers identified 37 individuals with CUL3 variants, most of which result in loss-of-function (LoF), leading to intellectual disabilities and possibly autistic traits; specific mechanisms affecting protein stability were also investigated.
  • - The findings enhance the understanding of NDDs associated with CUL3 mutations, suggesting that LoF variants are the main cause, which could help inform future diagnostics and treatment strategies.

Article Synopsis
  • - The study evaluates how different acclimatization times affect lung function measurements in cats with lower airway disease (LAD) using a noninvasive method called barometric whole-body plethysmography (BWBP).
  • - Twenty-four cats with LAD and eight healthy cats were tested, and significant variations in lung function metrics were observed between the first and third measurement periods, particularly in pause duration, peak inspiratory flow, and peak expiratory flow.
  • - The findings suggest that acclimatization time significantly impacts lung function measurements, indicating the need for potential adjustments in data analysis to improve accuracy in evaluating respiratory conditions in cats.

Cohort Expansion and Genotype-Phenotype Analysis of RAB11A-Associated Neurodevelopmental Disorder.

Pediatr Neurol

November 2024

Centre de recherche Azrieli du CHU Sainte-Justine, Montreal, Québec, Canada; Department of Pediatrics, University of Montreal, Montreal, Québec, Canada. Electronic address:

Article Synopsis
  • GTPases from the Rab family play a crucial role in membrane trafficking, and issues with these proteins have been linked to various neurological disorders, particularly involving RAB11A variants causing developmental and epileptic encephalopathy.
  • The study examined 16 patients with RAB11A variants, mostly de novo heterozygous missense mutations, finding that these variants are associated with intellectual disability, developmental delays, and a range of other physical and neurological symptoms.
  • The research suggests that while epilepsy is less common and less severe in patients with binding site mutations, the RAB11A neurodevelopmental disorder can affect multiple body systems, including gait, muscle tone, brain structure, and even fat distribution.

Article Synopsis
  • The study looks at how more young people are using new nicotine products and other products without nicotine, and how this could lead to serious health problems, suggesting that they should be banned.
  • Researchers checked a lot of research to see how these products affect young people's health.
  • Even though these products were thought to be safer, they can cause issues like asthma and heart problems, and they might make teens who never smoked start smoking, so health groups want them banned for kids.

Veterinarians rely on the measurement of canine body temperature to define the health status of dogs, but no studies exist defining a reference range for rectal temperature on a large group of dogs. The aim of this study was to define the rectal body temperature of dogs based on a large data set of diseased and healthy animals and to evaluate the capability of the employed algorithm to calculate reference intervals of numerical clinical data. Out of 24,013 recorded measurements, statistical analysis was applied to data from 9782 adult dogs that underwent clinical examination at a university clinic between 2008 and 2017.

Nephropathic cystinosis is an orphan autosomal recessive lysosomal storage disease characterized by a deficiency of cystinosin, a cystine transporter protein, leading to tissue damage, primarily in the kidney and cornea. With the introduction of cystine-depleting therapy with cysteamine and the possibility to survive to adulthood, new challenges of skeletal complications are a concern, with sparse data available regarding bone development. The aim of the current study was to gain more information on bone density and geometry in these patients.

Article Synopsis
  • Patients with a history of prior myocardial infarction (pMI) have worse outcomes compared to those without pMI when dealing with ischaemic heart failure with reduced ejection fraction (HFrEF).
  • In a study involving 2,075 patients, it was found that those with pMI experienced higher all-cause mortality, greater event rates for death or shock from an ICD, and a more significant decline in mobility and overall health status after one year.
  • These findings suggest that patients with ischaemic HFrEF and a history of pMI are at a higher risk, indicating a need for more specialized care for this subgroup.

A one-step synthesis of perylene dyes with lateral extension by condensed imidazoles in a cascade reaction of sodium amide and benzonitrile is described in which multiple extensions can be controlled by the reaction conditions. The extensions lead to bathochromic shifts in absorption and fluorescence while maintaining high fluorescence quantum yields. The condensed imidazole units cause additional absorption bands in the hypsochromic visible region, resulting in broad-band absorbers.
