MS-Based Glycome Characterization of Biotherapeutics With N- and O-Glycosylation.

Mass Spectrom Rev

January 2025

Asia-Pacific Glycomics Reference Site, Daejeon, Korea.

With the increasing FDA approvals of glycoprotein-based biotherapeutics including monoclonal antibodies, cytokines, and enzyme treatments, the significance of glycosylation in modulating drug efficacy and safety becomes central. This review highlights the crucial role of mass spectrometry (MS) in elucidating the glycome of biotherapeutics that feature N- and O-glycosylation, directly addressing the challenges posed by glycosylation complexity and heterogeneity. We have detailed the advancements and application of MS technologies including MALDI-TOF MS, LC-MS, and tandem MS in the precise characterization of glycoprotein therapeutics.

Background: Non-typhoidal Salmonella (NTS) frequently cause bloodstream infection in children under-five in sub-Saharan Africa, particularly in malaria-endemic areas. Due to increasing drug resistance, NTS are often not covered by standard-of-care empirical antibiotics for severe febrile illness. We developed a clinical prediction model to orient the choice of empirical antibiotics (standard-of-care versus alternative antibiotics) for children admitted to hospital in settings with high proportions of drug-resistant NTS.

The complex and dynamic nature of airborne fine particulate matter (PM) has hindered understanding of its chemical composition, sources, and toxic effects. In the first steps of a larger study, here, we aimed to elucidate relationships between source regions, ambient conditions, and the chemical composition in water extracts of PM samples ( = 85) collected over 16 months at an observatory in the Yellow Sea. In each extract, we quantified elements and major ions and profiled the complex mixtures of organic compounds by nontarget mass spectrometry.

In this paper, we propose a method to address the class imbalance learning in the classification of focal liver lesions (FLLs) from abdominal CT images. Class imbalance is a significant challenge in medical image analysis, making it difficult for machine learning models to learn to classify them accurately. To overcome this, we propose a class-wise combination of mixture-based data augmentation (CCDA) method that uses two mixture-based data augmentation techniques, MixUp and AugMix.

Background: Concurrent exercise (CE), an emerging exercise modality characterized by sequential bouts of aerobic (AE) and resistance exercise (RE), has demonstrated acute benefits on executive functions (EFs) and neuroelectric P3 amplitude. However, the effect of acute CE on inhibitory control, a sub-component of EFs, and P3 amplitude remains inconclusive. Moreover, exploring the mechanisms underlying the effects of acute exercise on EFs contributes to scientific comprehension, with lactate recognized as a crucial candidate positively correlated with EFs.

Cognitive training is a promising intervention for psychological distress; however, its effectiveness has yielded inconsistent outcomes across studies. This research is a pre-registered individual-level meta-analysis to identify factors contributing to cognitive training efficacy for anxiety and depression symptoms. Machine learning methods, alongside traditional statistical approaches, were employed to analyze 22 datasets with 1544 participants who underwent working memory training, attention bias modification, interpretation bias modification, or inhibitory control training.

Social cognition, which ranges from recognizing social cues to intricate inferential reasoning, is influenced by environmental factors and epigenetic mechanisms. Notably, methylation variations in stress-related genes like brain-derived neurotrophic factor (BDNF) and the oxytocin receptor (OXTR) are linked to distinct social cognitive functions and exhibit sex-specific differences. This study investigates how these methylation differences affect social cognition across sexes, focusing on both perceptual and inferential cognitive levels.

The increasing level of cadmium (Cd) contamination in soil due to anthropogenic actions is a significant problem. This problem not only harms the natural environment, but it also causes major harm to human health via the food chain. The use of chelating agent is a useful strategy to avoid heavy metal uptake and accumulation in plants.

Multi scale multi attention network for blood vessel segmentation in fundus images.

Sci Rep

January 2025

Department of Data Science and Artificial Intelligence, Sunway University, 47500, Petaling Jaya, Selangor Darul Ehsan, Malaysia.

Precise segmentation of retinal vasculature is crucial for the early detection, diagnosis, and treatment of vision-threatening ailments. However, this task is challenging due to limited contextual information, variations in vessel thicknesses, the complexity of vessel structures, and the potential for confusion with lesions. In this paper, we introduce a novel approach, the MSMA Net model, which overcomes these challenges by replacing traditional convolution blocks and skip connections with an improved multi-scale squeeze and excitation block (MSSE Block) and Bottleneck residual paths (B-Res paths) with spatial attention blocks (SAB).

To reliably operate anode-less solid-state Li metal batteries, wherein precipitated Li acts as the anode, stabilizing the interface between the solid electrolyte and electrode is crucial. The interface can be controlled by a metal interlayer on the electrolyte to form a Li alloy buffer that facilitates stable Li plating/stripping, thereby mitigating the loss of physical contact and preventing short circuits. However, the mechanism governing stable Li plating/stripping in the metal interlayer without degrading battery materials remains unclear owing to an incomplete understanding of the dynamic and complex electrochemical reactions in the solid state.

hESC-derived extracellular vesicles enriched with MFGE-8 and the GSH redox system act as senotherapeutics for neural stem cells in ischemic stroke.

Free Radic Biol Med

January 2025

Laboratory of Stem Cells and Tissue Regeneration, Department of Biotechnology, College of Life Sciences and Biotechnology, Korea University, Seoul 02841, South Korea. Electronic address:

Human embryonic stem cells (hESCs) and their extracellular vesicles (EVs) hold significant potential for tissue repair and regeneration. Neural stem cells (NSCs) in the adult brain often acquire senescent phenotypes after ischemic injuries, releasing neurodegenerative senescence-associated secretory phenotype factors. In this study, we investigated the senotherapeutic effects of hESC-EVs on NSCs and confirmed their neuroprotective effects in neurons via rejuvenation of NSC secretions.

The human lacrimal gland (LG), located above the outer orbital region within the frontal bone socket, is essential in maintaining eye surface health and lubrication. It is firmly anchored to the orbital periosteum by the connective tissue, and it is vital for protecting and lubricating the eye by secreting lacrimal fluid. Disruption in the production, composition, or secretion of lacrimal fluid can lead to dry eye syndrome, a condition characterized by ocular discomfort and potential eye surface damage.

January 2025

Department of Molecular Medicine and Biopharmaceutical Sciences, Graduate School of Convergence Science and Technology, Seoul National University, Gwanak-gu, Republic of Korea.

Although patients with anaplastic thyroid cancer (ATC) generally have a poor prognosis and there are currently no effective treatment options, survival and response to therapy vary between patients. Genomic and transcriptomic profiles of ATC have been reported; however, a comprehensive study of the tumor microenvironment (TME) of ATC is still lacking. This study aimed to elucidate the TME characteristics associated with ATC and their prognostic implications.
