Neuropeptide Y in cancer-biological functions and potential clinical implications.

Cancer Metastasis Rev

January 2025

Department of Biochemistry and Molecular and Cellular Biology, Georgetown University Medical Center, BSB 231A, 3900 Reservoir Rd., NW, Washington, DC, 20057, USA.

Neuropeptide Y (NPY) is a sympathetic neurotransmitter widely distributed in the peripheral and central nervous system, affecting many physiological functions. Consequently, dysregulation of the NPY system contributes to numerous pathological disorders, including stress, obesity, and cancer. The pleiotropic functions of NPY in humans are mediated by G protein-coupled receptors (Y1R, Y2R, Y5R), which activate several signaling pathways and thereby regulate cell growth, differentiation, apoptosis, proliferation, angiogenesis, and metabolism.

Isolated foot drop is a neurological sign frequently linked to lower motor neuron (LMN) lesions, including peroneal nerve damage or L4-L5 radiculopathy. Nonetheless, upper motor neuron (UMN) lesions, such as strokes or tumors located in the parasagittal motor cortex, may sometimes manifest as isolated foot drops. The main causes of isolated foot drop secondary to central etiologies are uncommon, with few instances documented in the literature.

  • Type 2 Diabetes Mellitus (T2DM) is a prevalent chronic condition in Saudi Arabia, and while bariatric surgery is known to aid in weight loss and potentially induce remission, long-term effects specific to the Saudi population are not well understood.
  • A retrospective study analyzed medical records of 74 Saudi adults who underwent bariatric surgery from 2016-2020, focusing on changes in Hemoglobin A1c (HbA1c) levels pre-and post-surgery and evaluating diabetes remission based on American Diabetes Association criteria.
  • Results showed significant improvements in glycemic control, with mean HbA1c levels dropping from 8.70 to 6.76 after one year; however, only 27%

Article Synopsis
  • The study investigates how magnesium (Mg) intake affects lipid profiles in patients with type 2 diabetes mellitus (T2DM), focusing on cholesterol and triglyceride levels.
  • Participants received nutritional education over three months to increase Mg-rich food consumption, resulting in a significant decrease in total cholesterol levels related to Mg intake.
  • The findings suggest that Mg-focused dietary education could be an effective, low-cost strategy for improving the health of T2DM patients, although further research is needed for long-term effects and larger populations.

Electronic address:

Four genes-DAND5, PKD1L1, MMP21, and CIROP-form a genetic module that has specifically evolved in vertebrate species that harbor motile cilia in their left-right organizer (LRO). We find here that CIROZ (previously known as C1orf127) is also specifically expressed in the LRO of mice, frogs, and fish, where it encodes a protein with a signal peptide followed by 3 zona pellucida N domains, consistent with extracellular localization. We report 16 individuals from 10 families with bi-allelic CIROZ inactivation variants, which cause heterotaxy with congenital heart defects.

EEFSEC deficiency: A selenopathy with early-onset neurodegeneration.

Institute of Medical Genetics and Applied Genomics, University of Tübingen, 72076 Tübingen, Germany; Center for Rare Disease, University of Tübingen, 72076 Tübingen, Germany; Genomics for Health in Africa (GHA), Africa-Europe Cluster of Research Excellence (CoRE).

Inborn errors of selenoprotein expression arise from deleterious variants in genes encoding selenoproteins or selenoprotein biosynthetic factors, some of which are associated with neurodegenerative disorders. This study shows that bi-allelic selenocysteine tRNA-specific eukaryotic elongation factor (EEFSEC) variants cause selenoprotein deficiency, leading to progressive neurodegeneration. EEFSEC deficiency, an autosomal recessive disorder, manifests with global developmental delay, progressive spasticity, ataxia, and seizures.
