Sortase A-mediated ligation (SML) or "sortagging" has become a popular technology to selectively introduce structurally diverse protein modifications. Despite the great progress in the optimization of the reaction conditions and design of miscellaneous C- or N-terminal protein modification strategies, the reported yields of conjugates are highly variable. In this study, we have systematically investigated C-terminal protein sortagging efficiency using a combination of several rationally selected and modified acceptor proteins and a panel of incoming surrogate non-peptidic amine nucleophile substrates varying in the structural features of their amino linker parts and cargo molecules.

Unlabelled: The study aims to investigate the clinicopathological significance of MRPL24 in human cancers, with a particular focus on breast cancer (BC). Comprehensive bioinformatics analyses were conducted using data from The Cancer Genome Atlas (TCGA) and various advanced database, including cBioPortal, UALCAN, TIMER, Prognoscan, TISIDB, KM Plotter, and The Human Protein Atlas, to provide a detailed evaluation of MRPL55's role in cancer. The findings were further validated through experimental studies.

The North Pontic region, which encompasses present-day Ukraine, was a crossroads of migration, connecting the vast Eurasian Steppe with Central Europe. We generated shotgun-sequenced genomic data for 91 individuals dating from around 7000 BCE to 1800 CE to study migration and mobility history in the region, with a particular focus on historically attested migrating groups during the Iron Age and the medieval period. We infer a high degree of temporal heterogeneity in ancestry, with fluctuating genetic affinities to different present-day Eurasian groups.

Crohn's disease (CD) and Ankylosing Spondylitis (AS) are chronic conditions with overlapping inflammatory pathways. This research investigates the genetic association between AS and the requirement for more aggressive therapeutic interventions in CD, suggesting a likelihood of increased severity in CD progression among individuals diagnosed with AS. This study utilized two-sample Mendelian randomization (TSMR) to analyze GWAS datasets for AS and CD requiring second-line treatment.

Material And Methods: Using multi-slice computed tomography (MSCT), the study examined the structure and relationships of key anatomical features, including the alveolar process, sinus wall thickness, and the position of the teeth in relation to the maxillary sinus.
Results: The results revealed that the lower wall of the maxillary sinus is predominantly formed by the alveolar process, with significant variability in wall thickness, especially between the central and lateral regions.
Human mesenchymal stromal cells (MSCs) are attractive for both medical practice and biomedical research. Nonfreezing short-term storage may provide safe and simple transportation and promote the practical use of MSCs. We aimed to determine the duration of efficient storage at ambient temperature (22°C) of human dermal MSCs in different three-dimensional organization and to investigate the role of cell metabolic mode in the resistance to the ambient storage damaging factors.

Patients And Methods: Materials and Methods: The study was carried out using a sociological method among palliative patients of three regions of Ukraine (Lviv, Kyiv and Kharkiv) on a random sample of 377 people surveyed during 2022-2024. The conducted research confirmed and clarified the previous results with greater reliability.

Background: Trends in the development of genetic markers for the purposes of genomic and marker-assisted selection primarily focus on identifying causative polymorphisms. Using these polymorphisms as markers enables a more accurate association between genotype and phenotype. Bioinformatic analysis allows calculating the impact of missense polymorphisms on the structural and functional characteristics of proteins, which makes it promising for identifying causative polymorphisms.

Vasculitis is a group of uncommon diseases characterized by inflammation of blood vessels, which contributes to the organ ischemia and damage. Cigarette smoke contains a high concentration of various toxins, which have the potential to affect the immune response and development of autoimmune/autoinflammatory rheumatic diseases including vasculitis. Smoking influences both innate and adaptive immune systems and plays binary functions in modulating immunity by either aggravating pathogenic immune responses or attenuating defensive immunity.

To date, only eight species of marine tardigrades have been recorded from the Southern Ocean. A total of 1210 tardigrade specimens were collected during various marine expeditions with R/V POLARSTERN: ANDEEP-1, ANDEEP-2, ANDEEP-3 and ANDEEP-SYSTCO. The sampled tardigrades belong to five families (Batillipedidae, Coronarctidae, Halechiniscidae, Styraconyxidae and Echiniscoididae), seven genera (Batillipes, Coronarctus, Moebjergarctus, Angursa, Styraconyx, Tholoarctus, Isoechiniscoides) and 15 species (Batillipes wyedeleinorum, Coronarctus dissimilis, Coronarctus tenellus, Coronarctus cf.

Nanostructured materials have been suggested to be used as a source of dietary zinc for livestock animals. In this study, we assessed the cytotoxicity of newly synthesized nanostructured zinc carbonate hydroxide (ZnCH) Zn(CO)(OH)microflakes. Cytotoxicity of the microflakes was assessed against murine L929 cell line and rat mature erythrocytes.

GdVO:Eu nanoparticles (OVNPs) have previously been shown to exhibit anti-aging effects in old rats. The accelerated aging model (overnutrition in early postnatal ontogenesis (POF)) was used to confirm the effect of OVNPs as a potential geroprotector. A comparative study of the effect of OVNPs, calorierestriction (CR) and CR-mimetic-metformin was carried out using a number of criteria: survival, prooxidant-antioxidant balance in the liver and blood, physiological parameters of male Wistar rats with accelerated aging.

Oral diseases are among the most common non-communicable diseases around the globe and become a public health challenge that considerably impact general health and well-being in all ages across the lifespan. Oral health is an integral part of general health, where poor oral health and other lifestyle-related chronic diseases have a common risk factor background. The underlying causes of oral health inequalities are often complex and related to country-specific historical, economic, cultural, social, or political factors.

This research focused on the helicopter turboshaft engine dynamic model, identifying task solving in unsteady and transient modes (engine starting and acceleration) based on sensor data. It is known that about 85% of helicopter turboshaft engines operate in steady-state modes, while only around 15% operate in unsteady and transient modes. Therefore, developing dynamic multi-mode models that account for engine behavior during these modes is a critical scientific and practical task.

Background: Following the 2022 Russian invasion, Ukraine's healthcare system suffered extensive damage, with over 1000 medical facilities destroyed, exacerbating the trauma care crisis. The absence of standardized trauma training left Ukrainian healthcare providers ill-equipped to manage the surge in trauma cases amid conflict. To bridge this gap, we implemented advanced trauma life support (ATLS) courses in Ukraine amid active warfare, aiming to enhance trauma care expertise among healthcare professionals.

Introduction: Hyperpigmentation disorders are caused by excess production of the pigment melanin, catalyzed by the enzyme tyrosinase. Novel tyrosinase inhibitors are needed as therapeutic agents to treat these conditions.
Method: To discover new inhibitors, we performed a virtual screening of the ZINC20 library containing 1.
