Prostate cancer antigen 3 (PCA3) has emerged as a critical biomarker for the early detection of prostate cancer, complementing the traditional prostate-specific antigen (PSA) testing. This research presents a novel resistive sensor based on reduced graphene oxide (RGO) functionalized with glutaraldehyde (GA)/complementary single-stranded DNA (ss-DNA) for the detection of the PCA3 RNA. The device was meticulously characterized at each fabrication step to confirm the successful integration of the various layers on the sensor device, utilizing atomic force microscopy (AFM) which confirmed the increase in the thickness of the sensor from ∼1.

Introduction: The dearth of population-based data on polysubstance use, especially in young emerging adults, presents a critical gap in understanding public health challenges, particularly in rural India, where tobacco and oral cancer are prevalent. This study addresses this gap by investigating the prevalence and patterns of substance use, with a focus on young emerging adults (18-29 years) within a high-risk cohort for oral cancer in Varanasi, India.

Methods: The study involved a cross-sectional analysis of baseline data from 10,101 participants within a high-risk cohort in rural settings of the Varanasi district between December 2020-June 2023.

This article is part of the Dendritic Cell Guidelines article series, which provides a collection of state-of-the-art protocols for the preparation, phenotype analysis by flow cytometry, generation, fluorescence microscopy, and functional characterization of mouse and human dendritic cells (DC) from lymphoid organs, and various nonlymphoid tissues. Within this article, detailed protocols are presented that allow for the generation of single-cell suspensions from human nonlymphoid tissues including lung, skin, gingiva, intestine as well as from tumors and tumor-draining lymph nodes with a subsequent analysis of dendritic cells by flow cytometry. Further, prepared single-cell suspensions can be subjected to other applications including cellular enrichment procedures, RNA sequencing, functional assays, etc.

Background: Polysubstance dependence has emerged as a clinically distinct entity from monosubstance dependence, yet its impact on patients' wives is understudied. Wives of substance-dependent men face a tremendous burden in their social lives, leading to faulty coping and burden of care. Comparative studies between monosubstances and polysubstances are lacking.

Aim: Older patients with locally advanced head and neck cancers (LA-HNC) are under-represented in clinical trials and denied standard treatment with concurrent chemo-radiation. Most are treated with radiotherapy (RT) alone. However, with the use of Intensity Modulated Radiation Therapy (IMRT) and good supportive care, even this cohort of patients can be considered for chemo-radiation.

In the era of free flaps, we propose a simple yet effective local flap, the digastric muscle flap, to reconstruct floor of the mouth defects and to repair an orocervical fistula that is created during excision of tongue and/or floor of the mouth malignancies. The digastric muscle flap was used in 15 patients who were diagnosed with oral squamous cell carcinoma of the tongue and/or floor of the mouth. Partial glossectomy was performed in 4 cases, hemiglossectomy in 4 cases, and wide local excision in 7 cases.

August 2024

Department of Gastroenterology and Hepatology, Karnatak Lingayat Education Society's (KLES) Dr. Prabhakar Kore Hospital and Medical Research Centre, KLE Academy of Higher Education and Research, Jawaharlal Nehru Medical College, Belagavi, IND.

Background: India is currently facing an epidemic of type 2 diabetes mellitus (T2DM) and obesity which are high-risk factors for the development of non-alcoholic fatty liver disease (NAFLD). A non-invasive tool, the vibration-controlled transient elastography (VCTE; FibroScan, Echosens, Paris, France) is used to diagnose NAFLD.

Aim: To identify the prevalence, spectrum, and metabolic determinants of NAFLD in Indian adult women using liver function tests (LFT) and non-invasive FibroScan (liver stiffness measure, i.

Department of Haematogenetics, ICMR-National Institute of Immunohematology, Indian Council of Medical Research, 13th Floor, NMS Building, King Edward Memorial (KEM.) Hospital Campus, Parel, Mumbai, 400012, India.

Article Synopsis
  • * The study involves identifying CSA patients using a targeted next-generation sequencing (t-NGS) panel, discovering mutations related to microcytic anaemia, and assessing their response to treatments such as pyridoxine and blood transfusions.
  • * Researchers found various genetic variants among patients, primarily in European populations, and used PyMoL software to study how these mutations affect protein structure and function, aiming for a deeper understanding of CSA genetics.

Article Synopsis
  • Rare disorders include about 7,500 different conditions that are challenging to diagnose due to a lack of specialized healthcare, testing facilities, and treatment options, particularly in countries like India with diverse population groups.
  • This study examined a cohort of 3,294 patients with 305 identified rare genetic diseases, primarily affecting the neuromuscular and neurodevelopmental systems, as well as inborn errors of metabolism.
  • The findings revealed that the most common diseases were Gaucher disease in the IEM category, and Duchenne muscular dystrophy and trinucleotide repeat expansion disorders in the NMND group, highlighting the need for more focused genetic research and healthcare resources in India.

Article Synopsis
  • Current diagnostic methods for lysosomal storage disorders (LSDs) in India are lengthy and expensive, relying on biochemical tests and DNA sequencing, which often yield low results due to overlapping symptoms.
  • Researchers have created a novel, cost-effective sequencing assay using single-molecule molecular inversion probes (smMIPs) that accurately identifies genetic variants linked to 29 common LSDs.
  • The new assay showed a high diagnostic yield of 83.4% in patients with previous biochemical diagnoses and effectively detected rare diseases like Niemann-Pick type C, outperforming traditional methods and allowing for flexible use with different sample types.
