Understanding the pathogenesis mechanism of the Monkeypox virus (MPXV) is essential to guide therapeutic development against the Monkeypox virus. In the current study, we investigated the impact of the only two reported substitutions, S30L, D88N, and S30L-D88N on the G9R of the replication complex in 2022 with E4R using structural modeling, simulation, and free energy calculation methods. From the molecular docking and dissociation constant (K) results, it was observed that the binding affinity did not increase in the mutants, but the interaction paradigm was altered by these substitutions.

Background: Inherited bleeding, thrombotic, and platelet disorders (BTPDs) are a heterogeneous set of diseases, many of which are very rare globally. Over the past 5 decades, the genetic basis of some of these disorders has been identified, and recently, high-throughput sequencing has become the primary means of identifying disease-causing genetic variants.
Objectives: Knowledge of the clinical validity of a gene-disease relationship is essential to provide an accurate diagnosis based on results of diagnostic gene panel tests and inform the construction of such panels.

Background And Objectives: Declines in stroke admission, IV thrombolysis (IVT), and mechanical thrombectomy volumes were reported during the first wave of the COVID-19 pandemic. There is a paucity of data on the longer-term effect of the pandemic on stroke volumes over the course of a year and through the second wave of the pandemic. We sought to measure the effect of the COVID-19 pandemic on the volumes of stroke admissions, intracranial hemorrhage (ICH), IVT, and mechanical thrombectomy over a 1-year period at the onset of the pandemic (March 1, 2020, to February 28, 2021) compared with the immediately preceding year (March 1, 2019, to February 29, 2020).

Introduction: The inferior alveolar nerve (IAN) can occasionally be observed in the extraction socket of the mandibular third molar (M3M) intraoperatively. Exposure of inferior alveolar neurovascular bundle during surgery primarily depends upon the absence of bony cortex between the canal and root of impacted third molar or either by existence of a very thin cortical lining between two which gets broken during luxation of tooth. Accurate anatomical relationship of inferior alveolar canal with root apex of impacted (M3M) and the location of canal can be determined by Cone beam computed tomography (CBCT).

SARS-CoV-2, an RNA virus, has been prone to high mutations since its first emergence in Wuhan, China, and throughout its spread. Its genome has been sequenced continuously by many countries, including Pakistan, but the results vary. Understanding its genomic patterns and connecting them with phenotypic features will help in devising therapeutic strategies.

Aquaporin 4 (AQP4) is a water transporting, transmembrane channel protein that has important regulatory roles in maintaining cellular water homeostasis. Several other AQP proteins exhibit calmodulin (CaM)-binding properties, and CaM has recently been implicated in the cell surface localization of AQP4. The objective of the present study was to assess the CaM-binding properties of AQP4 in detail.

Objective: The goal of this systematic review and meta-analysis is to determine the performance of 4% Articaine 2% Lidocaine for mandibular and maxillary block and infiltration anaesthesia in patients with irreversible pulpitis (IP).
Methods: PubMed/MEDLINE, Cochrane Central Register of Controlled Trials, Web of Science, Google Scholar, and Open Gray were used to conduct a thorough literature search. A manual search of the reference lists of the publications found was also carried out.

Background: The aim of the study was to evaluate the effect of 10% alpha-tocopherol and 5% grape seed extract on the microhardness and shear bond strength (SBS) to bleached human dentin.
Materials And Methods: This study was done on 200 extracted premolars which were decoronated and grinded to get flat dentin surface occlusaly. They were divided into four groups: (a) bleaching, (b) bleaching and application of alpha-tocopherol, (c) bleaching and application of grape seed extract, and (d) control.

Aims: The purpose of the study was to determine the best combination for chemomechanical preparation in primary teeth using two endo file systems (hand and rotary) along with two different irrigants.
Materials And Methods: Sixty primary molars indicated for pulpectomy, underwent chemomechanical preparation using endo files (H hand files and rotary Protaper files) and root canal irrigating solutions (Smearclear and QMiX). Samples from root canals were collected before and after the chemomechanical preparation of the canal and were cultured for microbial analysis of .

When endodontically diagnosing and evaluating the morphology and anatomy of roots and root canals, especially in multirooted teeth, clinicians must exercise extremely keen vision and give careful attention to minute details. Fundamental knowledge of the most common anatomic characteristics and the possible variations of these characteristics is necessary to provide successful endodontic therapy. While all mandibular molars have significant variations in roots and root canal configuration, mandibular second molars perhaps present the biggest challenge.

Background: Thyroid peroxidase antibodies are associated with an increased risk of miscarriage and preterm birth, even when thyroid function is normal. Small trials indicate that the use of levothyroxine could reduce the incidence of such adverse outcomes.
Methods: We conducted a double-blind, placebo-controlled trial to investigate whether levothyroxine treatment would increase live-birth rates among euthyroid women who had thyroid peroxidase antibodies and a history of miscarriage or infertility.

Aim: This study aimed to evaluate the effect of cigarette smoke on the surface roughness of two commercially available denture base materials.
Materials And Methods: A total numbers of 40 specimens were fabricated from two commercially available denture base materials: heat-cured polymethylmethacrylate and flexible denture base materials (20 for each). The specimens for each type were divided into four subgroups: subgroup I, heat-cured denture base material specimens (control group); subgroup II, flexible denture base material (control group); subgroup III, heat-cured denture base material specimen exposed to cigarette smoking group; and subgroup IV, flexible denture base material specimens exposed to cigarette smoking.
