Mitochondrial fragmentation in cigarette smoke-induced bronchial epithelial cell senescence.

Am J Physiol Lung Cell Mol Physiol

November 2013

Division of Respiratory diseases, Dept. of Internal Medicine, Jikei Univ. School of Medicine, Japan.

Article Synopsis
  • Mitochondria constantly change shape through fission and fusion, and disruptions in this process can lead to increased production of reactive oxygen species (ROS), contributing to diseases like COPD.
  • The study focused on how cigarette smoke extract (CSE) affects mitochondrial dynamics and ROS production, leading to cellular senescence in human bronchial epithelial cells (HBECs).
  • Findings show that CSE induces mitochondrial fragmentation and ROS production in HBECs, accelerating cellular senescence, but this effect can be mitigated by antioxidants.

Laparoscopic surgery has been used for various areas of abdominal cavity. However, the laparoscope has limitation of its field of view and mobility. Our aim is to develop a new video camera system that acquires multiple viewpoints in the abdominal cavity.

Within the digestive organ surgery robot R&D project, our research team aims to develop a surgical robot training device with an interface identical to that of the actual device. The training device uses an organ model that changes shape in real time to train operators to grab, cut open, and cut off soft tissues and close wounds using the actual device. To increase the effectiveness of the training device, we added functions to save the movements of the robot in training and changes in the operation field.

Reepithelialization of remodeled air spaces with bronchial epithelial cells is a prominent pathological finding in idiopathic pulmonary fibrosis (IPF) and is implicated in IPF pathogenesis. Recent studies suggest that epithelial senescence is a risk factor for development of IPF, indicating such reepithelialization may be influenced by the acceleration of cellular senescence. Among the sirtuin (SIRT) family, SIRT6, a class III histone deacetylase, has been demonstrated to antagonize senescence.

Endothelin-1 (ET-1) shows a positive inotropic effect on cardiac muscle. Although the L-type Ca(2+) current (I(Ca)) is one of the important determinants of cardiac excitation-contraction coupling, the effect of ET-1 on the I(Ca) is not always clear. The controversial results appear to be due to different patch-clamp methods.

In this study we have designed and created a data-fusion display that has enabled volumetric MIP image navigation using intraoperative C-arm CT data in the operating room. The 3D volumetric data reflecting a patient's inner structure is directly displayed on the monitor through video images of the surgical field using a 3D optical tracking system, a ceiling-mounted articulating monitor, and a small size video camera mounted at the back of the monitor. The system performance was validated in an experiment carried out in the operating room.

Intra-operative navigation in which the target position is provided to assist an intuitive understanding of the surgical field has been studied and applied in many clinical areas. Position measurement of a surgical field is usually performed with a magnetic sensor, a marker type optical position sensor. For navigation of hard tissue, the measurement of several markers dispersedly located on the surface is enough to detect the position of an object that can be assumed as a rigid body.

Between April 1991 and August 1992, we diagnosed 51 cases of CFS who met definition of CFS designated by CDC, 1988. They are 41 female and 11 male, and 78% are women. At first visit, their ages are ranged from 16 to 64 years old, and approximately 45% is 20 to 30 years old.

This article reviewed Definition of CFS proposed by CDC 1988. There are several issues in Definition for CFS of CDC. It is presented that other chronic clinical conditions have been satisfactorily excluded, including preexisting psychiatric diseases in (2) of major criteria.
