Article Synopsis
  • Accurate assessment of left ventricular filling pressure in patients with atrial fibrillation is challenging, but a new scoring system called VMT score can visually assess valve opening timings to evaluate this pressure.
  • In a study with 119 patients, the VMT score showed a strong correlation with elevated pulmonary arterial wedge pressure and had a diagnostic accuracy of 87%, outperforming traditional Doppler methods.
  • The findings were validated in a separate group of 189 patients, confirming that a VMT score of 2 or higher indicates significant left ventricular filling pressure elevation, making it a potentially valuable tool in clinical settings.

Department of Hematology and Oncology, Graduate School of Medicine, Faculty of Medicine, Osaka University, Osaka 565-0871, Japan.

The advent of recycling antibodies, leveraging pH-dependent antigen binding and optimized FcRn interaction, has advanced the field of antibody therapies, enabling extended durability and reduced dosages. Eculizumab (Soliris) demonstrated the efficacy of C5 inhibitors for paroxysmal nocturnal hemoglobinuria (PNH), while its derivative, ravulizumab (Ultomiris), recognized as a recycling antibody, extended the dosing intervals. However, limitations including intravenous administration and inefficacy in patients with the R885H single-nucleotide polymorphism (SNP) in C5 could necessitate alternative solutions.

Targeting Oxidative Phosphorylation with a Novel Thiophene Carboxamide Increases the Efficacy of Imatinib against Leukemic Stem Cells in Chronic Myeloid Leukemia.

Int J Mol Sci

October 2024

Division of Hematology, Respiratory Medicine and Oncology, Department of Internal Medicine, Faculty of Medicine, Saga University, Saga 849-8501, Japan.

Patients with chronic myeloid leukemia (CML) respond to tyrosine kinase inhibitors (TKIs); however, CML leukemic stem cells (LSCs) exhibit BCR::ABL kinase-independent growth and are insensitive to TKIs, leading to disease relapse. To prevent this, new therapies targeting CML-LSCs are needed. Rates of mitochondria-mediated oxidative phosphorylation (OXPHOS) in CD34CML cells within the primitive CML cell population are higher than those in normal undifferentiated hematopoietic cells; therefore, the inhibition of OXPHOS in CML-LSCs may be a potential cure for CML.
