Article Synopsis
  • Abruptly stopping heavy cannabis use can lead to withdrawal symptoms like irritability, anxiety, insomnia, and in some cases, trigger acute psychosis.
  • A systematic review and a comprehensive analysis of psychiatric records identified a total of 112 individuals whose first episodes of psychosis were linked to cannabis withdrawal.
  • Continued cannabis use after experiencing a psychotic episode significantly increases the risk of relapse compared to individuals who choose to abstain.

Article Synopsis
  • - The study investigates how bromoform (CHBr) behaves when exposed to near-infrared (NIR) light, focusing on its breakdown and isomerization in cationic states through advanced imaging techniques.
  • - It is observed that the dissociation process, particularly when forming HBr and Br fragments, occurs with a delay compared to the faster breakdown of the C-Br bond.
  • - Molecular dynamics simulations indicate that this delay results from temporary isomerization processes involving H- and Br-migrations before the final fragments are produced.

Adolescent urinary concentrations of phthalate metabolites and indices of overweight and cardiovascular risk in Dutch adolescents.

Environ Int

December 2024

Division of Neonatology, Department of Pediatrics, Beatrix Children's Hospital, University Medical Center Groningen, University of Groningen, Hanzeplein 1, PO Box 30.001, 9713 GZ Groningen, the Netherlands.

Phthalates have been linked to cardiovascular risk factors. Exposure to chemicals with endocrine disrupting properties during the pubertal period can interfere with normal endocrine processes. This study aims to determine whether adolescent urinary concentrations of phthalate metabolites are associated with indices of overweight and cardiovascular risk in 13-15-year-old children.

Ethylene glycol (EG) is a versatile molecule produced in the petrochemical industry and is widely used to manufacture plastic polymers, anti-freeze, and automotive fluids. Biotechnological production of EG from xylose, a pentose present in lignocellulose biomass hydrolysates, has been achieved by the engineering of bacteria, such as Escherichia coli and Enterobacter cloacae, and the yeast Saccharomyces cerevisiae with synthetic pathways. In the present work, the Dahms pathway was employed to construct Komagataella phaffii strains capable of producing EG from xylose.

Consensus on the key characteristics of metabolism disruptors.

Nat Rev Endocrinol

November 2024

Department of Developmental and Cell Biology, University of California, Irvine, Irvine, CA, USA.

Metabolism-disrupting agents (MDAs) are chemical, infectious or physical agents that increase the risk of metabolic disorders. Examples include pharmaceuticals, such as antidepressants, and environmental agents, such as bisphenol A. Various types of studies can provide evidence to identify MDAs, yet a systematic method is needed to integrate these data to help to identify such hazards.

Purpose: To investigate the predictive value of RECIST response within 3, 6, or 12 months on long-term survival, and explore differences between nivolumab+ipilimumab and nivolumab monotherapy, we analyzed pooled 5-year data of 935 responder and non-responder patients at various time points after treatment initiation in CheckMate 069, 066, and 067 studies.

Patients And Methods: Treatment-naive advanced melanoma patients received nivolumab+ipilimumab or nivolumab monotherapy. To decrease immortal time bias, 3-, 6-, or 12-month overall survival (OS) and progression-free survival (PFS) landmark analyses were performed.

Congenital and Infantile Masses of the Head and Neck.

Radiographics

December 2024

From the Department of Radiology, Monroe Carell Jr Children's Hospital at Vanderbilt, 2200 Children's Way, Nashville, TN 37232 (A.M.F., R.K., E.S., D.M., K.D.C., L.J., S.P., A.S.); and Department of Radiology, Harvard University, Boston Children's Hospital, Boston, Mass (C.D.R., J.R.).

The spectrum of congenital and infantile masses of the head and neck is broad, including developmental and neoplastic entities. The diseases encountered in this vulnerable patient population differ substantially from those in older children and adults. Familiarity with the types of encountered masses, typical imaging characteristics, and expected clinical course is critical for radiologists who care for pregnant women (fetuses) and infants.

  • To prevent HIV-1 transmission, high levels of broadly neutralizing antibodies are necessary at mucosal sites of exposure, particularly in the colorectal and genitourinary tracts.
  • A study compared the biodistribution of two monoclonal antibodies, VRC01 and its longer-lasting variant VRC01LS, over 1-52 weeks post-infusion, finding VRC01LS levels significantly higher in various tissues at earlier and later time points.
  • While both antibodies are mainly retained in rectal and cervical tissue, only a small percentage reaches seminal and rectal secretions; VRC01LS shows a longer elimination half-life, indicating its potential for sustained protection against HIV-1.

  • Antidepressants are commonly prescribed to older adults for depression, but their impact on recovery after hip fractures is not well understood.
  • A study involving nearly 18,000 older adults with depression who underwent hip fracture surgery found that those using antidepressants before the injury had shorter hospital stays.
  • The findings suggest that antidepressant use does not hinder recovery post-hip fracture and may even be beneficial, with no significant differences based on sex noted in the results.

Nanopore sequencing of intact aminoacylated tRNAs.

bioRxiv

November 2024

University of Colorado School of Medicine, Department of Biochemistry and Molecular Genetics, RNA Bioscience Initiative, Aurora, Colorado.

Transfer RNAs (tRNA) are decorated during biogenesis with a variety of modifications that modulate their stability, aminoacylation, and decoding potential during translation. The complex landscape of tRNA modification presents significant analysis challenges and to date no single approach enables the simultaneous measurement of important but disparate chemical properties of individual, mature tRNA molecules. We developed a new, integrated approach to analyze the sequence, modification, and aminoacylation state of tRNA molecules in a high throughput nanopore sequencing experiment, leveraging a chemical ligation that embeds the charged amino acid in an adapted tRNA molecule.

Article Synopsis
  • This study explores the relationships between OCT biomarkers and contrast sensitivity in eyes diagnosed with intermediate age-related macular degeneration (iAMD), aiming to improve understanding of disease progression.
  • A total of 205 eyes from 134 patients were analyzed; findings reveal that higher retinal pigment epithelium volume and intraretinal hyperreflective foci correlate with poorer contrast sensitivity measures.
  • Over a follow-up period, approximately 43% of eyes progressed to late-stage AMD, indicating the potential clinical significance of the identified biomarkers in predicting progression.

Observations of maxillary (upper bill) bending in hummingbirds have been considered an optical illusion, yet a recent description of out-of-phase opening and closing between their bill base and tip suggests a genuine capacity for bill bending. We investigate bill kinematics during nectar feeding in six species of hummingbirds. We employed geometric morphometrics to identify bending zones and combined these data with measurements of bill flexural rigidity from micro-computed tomography scans to better understand the flexing mechanism.
