The gtfT gene and its upstream region isolated from the Streptococcus sobrinus OMZ176 chromosomal DNA were sequenced. The gtfT gene was preceded by a potential Shine-Dalgarno sequence. The gtfT gene product, glucosyltransferase (GTF), displays a typical gram-positive bacterial signal peptide sequence and both an active site peptide sequence and carboxy-terminal repeats typical of GTFs.

The effect of androgen on protein kinase activities was studied in chromosomal proteins from female mouse submandibular gland. The protein kinase activities in the nuclei were stimulated 3 h after testosterone administration. The in vitro addition of cAMP in the nuclei resulted in the enhancement of the androgen-sensitive protein kinase activities.

The cytochemical localization of Ca(++)-ATPases in Meissner's corpuscles of the palatine mucosa of the Mongolian gerbil was studied. Wachstein-Meisel's medium modified by SALAMA et al. (1987) was used for the demonstration of low-affinity Ca(++)-ATPase or Ca++/Mg(++)-ATPase, whereas the medium devised by Körtje et al.

An N-terminal truncated androgen receptor with putative DNA- and ligand-binding domains (AR438) and that with a ligand-binding domain (AR612) were expressed under control of the T7 promoter in E. coli or translated in vitro with rabbit reticulocyte lysate, and their ligand-binding properties and the interaction with HSP90 were investigated. Bacterially expressed AR438 and AR612 bound a synthetic androgen, [3H]R1881, with apparent dissociation constants of 2.

To examine the ultimate fate of hypertrophic chondrocytes, Meckel's cartilage bars from 18-day-old mouse embryos were transplanted into isogenic mouse spleen for 3, 7, 14 and 21 days and observed at the light and electron microscopic levels. The midportions of these Meckel's cartilage bars were used as explants; they were characterized by many hypertrophic chondrocytes containing euchromatic round nuclei, a large amount of glycogen particles, and some vacuoles. Grafted cartilage adapted well to the splenic tissue, showing intense metachromasia around the territorial matrix.

Resting cartilages taken from 2-day-old mouse ribs were transplanted into spleens in order to carry out morphological investigations of the life cycles of their chondrocytes. The explants were isografted for periods of up to 60 days and examined at light and electron microscopic levels, using von Kossa's reaction or osmium-potassium ferrocyanide (OPF) fixation. By day 3 after transplantation, resting cartilage containing immature chondrocytes was well adapted to splenic tissue and by 7 days after transplantation these chondrocytes had changed into early hypertrophic chondrocytes containing large vacuoles, glycogen aggregates and abundant secretory organelles.

Retinoic acid (RA) binding has been detected in the nuclei of a subclone (CL-1) of human submandibular adenocarcinoma cell line HSG conditioned to grow in a serum-free defined medium. Competition assay confirmed the specificity of the RA binding. Scatchard analysis showed the binding molecule to have a high affinity and low capacity.

The gtf gene coding for glucosyltransferase (GTF), which produces a water-soluble glucan, was cloned from Streptococcus sobrinus OMZ176 (serotype d) into plasmid vector pBR322. This gene was expressed in Escherichia coli, and the product was purified to near homogeneity. The antigenicity of recombinant GTF (rGTF) was examined with the antisera raised against purified GTF P1, P2, P3, and P4 obtained from S.

The present study was designed to demonstrate the short term mineralization of dentin and enamel, and to investigate the effects of sodium beta-glycerophosphate (Na-beta-GPO4) on calcification in a serum-free, chemically-defined medium. The first mandibular molars at the bell stage dissected from 18-day-old mouse embryos were used as explants, which were cultured by an improved flotation method. Calcification of enamel in the 18-day-old embryonic molars occurred within the 6th day of culture.

Transformed and bacterially expressed glucocorticoid receptors free from Mr 90,000 heat shock protein (hsp90) have a 100-fold lower steroid-binding affinity than the hsp90-bound nontransformed receptor, suggesting that hsp90 is needed for high-affinity steroid binding [Nemoto, T., Ohara-Nemoto, Y., Denis, M.

Human salivary gland adenocarcinoma (HSG) cells treated with 10(-6) M triamcinolone acetonide for 48 h exhibited a 1.7- to 2.0-fold increase in [125I]human epidermal growth factor (hEGF) binding capacity as compared with untreated HSG cells.

We found that when the midsection of Meckel's cartilage bars obtained from mice on the eighteenth day of gestation were grafted into isogenic mouse spleen, chondrocytes induced an endochondral calcification. Concurrent with the onset of calcification throughout Meckel's cartilage matrix, periodic banded thick collagen fibrils and matrix vesicles were observed around the chondrocytes. Although most of the chondrocytes prior to grafting were hypertrophic cells, they survived for seven days in the splenic tissue and had well-developed secretory organelles.

A ganglionated nerve plexus (GNP) found in the nasal vestibular mucosa of the adult musk shrew, and most likely belonging to the nervus terminalis, was studied by electron microscopy. The GNP, which contained ganglion cells and both myelinated and unmyelinated nerve fibers, was located in the subepithelial layer of the mesial vestibular mucosa of the snout processes. The ganglion cells were characterized by an eccentrically located round nucleus, dispersed mitochondria and Golgi apparatuses, and many small vesicles.

Mandibular defects in six patients were immediately reconstructed with pure titanium reconstruction plates and autogenous iliac bone blocks. However, an alveolar process adequate for functional fitting of a denture did not form. More than 6 months after the reconstruction, correction of the ridge deficiency was attempted using autogenous particulate iliac cancellous bone and marrow combined with hydroxylapatite particles.

A nation-wide field survey was carried out to investigate the etiology of enamel hypoplasia for permanent teeth in 286 subjects below 20 years of age with permanent dentition having enamel hypoplasia. Visual examination, photographic evaluation and questionnaire survey were used. The same questionnaire survey was also carried out in 111 subjects without hypoplastic teeth as controls.

Human salivary gland adenocarcinoma cell line HSG secretes an epidermal growth factor (EGF)-like molecule and contains EGF receptors. Growth of HSG cells is inhibited by glucocorticoid. We have identified that the growth inhibition by glucocorticoid is induced by the reduced secretion of the EGF-like molecule and that addition of anti-human EGF antibody to the culture specifically inhibits the growth of HSG cells, suggesting that autocrine secretion is involved in the growth of HSG cells.

Mandibular first molars in mice ranging in age from 18 days prenatal to 5 days postnatal were used for light and electron microscopic examinations of the enamel-free area (EFA) during development of the occlusal cusp (mesiobuccal cusp). Notable morphological changes in the inner enamel epithelium and the cells of the stratum intermedium were observed. At prenatal age of 18 days, the inner enamel epithelium of the EFA (EFA epithelium) was composed of a layer of columnar cells and covered by the cells of the stratum intermedium.

Lamellated sensory corpuscles in the perioral tissues of the adult cat and the adult miniature pig were studied by light and electron microscopy. In the cat lip, over half of the lamellated corpuscles existed in an aggregate form associated with other nerve elements and blood capillaries in peripheral nerve swellings, while the remainder existed in an isolated form. Conversely, almost all the lamellated corpuscles in the miniature pig existed in an aggregate form in nerve swellings.
