Fabry disease (FD) is a rare disorder resulting from a genetic mutation characterized by the accumulation of sphingolipids in various cells throughout the human body, leading to progressive and irreversible organ damage, particularly in males. Genetically-determined deficiency or reduced activity of the enzyme (alpha - Galactosidase; α-Gal) leads to the accumulation of sphingolipids in the lysosomes of various cell types, including the heart, kidneys, skin, eyes, central nervous system, and digestive system, triggering damage, leading to the failure of vital organs, and resulting in progressive disability and premature death. FD diagnostics currently depend on costly and time-intensive genetic tests and enzymatic analysis, often leading to delayed or inaccurate diagnoses, which contribute to rapid disease progression.

Background: Streptomyces roseochromogenes NRRL 3504 produces clorobiocin, an aminocoumarin antibiotic that inhibits DNA replication. No other natural products have been isolated from this bacterium so far, despite the presence of a rich repertoire of specialized metabolite biosynthesis gene clusters (smBGCs) within its genome. Heterologous expression of smBGCs in suitable chassis speeds up the discovery of the natural products hidden behind these sets of genes.

Spectroscopic properties of Tb-doped and Tb-Ag codoped lithium tetraborate (LTB) glasses with LiBO (or LiO-2BO) composition are investigated and analysed using electron paramagnetic resonance (EPR), optical absorption, photoluminescence (PL) and photoluminescence excitation (PLE) spectra, PL decay kinetics and absolute quantum yield (QY) measurements. PL spectra of the investigated glasses show numerous narrow emission bands corresponding to the D → F (J = 6-0) and D → F (J = 5-3) transitions of Tb (4f) ions. The most intense PL band of Tb ions at 541 nm (D → F transition) is characterised by a lifetime slightly exceeding 2.

Colorectal cancer is influenced by genetic mutations, lifestyle factors, and diet, particularly high fat intake, which raises bile acid levels in the intestinal lumen. This study hypothesized that bile acids contribute to tumorigenesis by disrupting ion transport and ATPase activity in the intestinal mucosa. The effects of 3-sulfo-taurolithocholic acid (TLC-S) on ATPase activity were investigated in colorectal cancer samples from 10 patients, using adjacent healthy tissue as controls, and in rodent liver function.

In this article, we provide an overview of hydrogen storage materials, taking our previous results as examples. Towards the end of the paper, we present a case study in order to highlight the effects of substitutional alloying, compositional additives, and nanostructuring on the hydrogen sorption properties of magnesium-based intermetallics. Specifically, partial substitution of Mg by Li and d-elements by p-elements leads to structural changes, inducing disorder and the formation of high-entropy alloys.

A series of novel spirocyclic α-proline building blocks with a spiro conjunction in position 3 of the pyrrolidine ring was prepared to employ two convenient and practical synthetic approaches. Both alternative routes utilize simple and readily available starting materials─cyclic ketones and esters─and comprise 6 and 7 steps, respectively. The methodologies feature distinct advantages, using routine organic chemistry transformations, and are suitable for producing multigram amounts of the target prolines.

Specialized or secondary metabolites are small molecules of biological origin, often showing potent biological activities with applications in agriculture, engineering and medicine. Usually, the biosynthesis of these natural products is governed by sets of co-regulated and physically clustered genes known as biosynthetic gene clusters (BGCs). To share information about BGCs in a standardized and machine-readable way, the Minimum Information about a Biosynthetic Gene cluster (MIBiG) data standard and repository was initiated in 2015.

The previously neglected "angular" spirocyclic azetidines have been synthesized, characterized, and validated in drug discovery. We have shown that these compounds could act as bioisosteres for common saturated six-membered heterocycles. Their incorporation into the structure of the anticancer drug Sonidegib (instead of morpholine), and Danofloxacine (instead of piperazine) provided novel patent-free analogs with similar physicochemical properties and high activity.

To enhance the usually low-charge carrier mobilities of highly twisted donor-acceptor-type compounds that exhibit thermally activated delayed fluorescence, we designed a rodlike acceptor benzodioxinoquinoxaline. This acceptor and two donor-acceptor-donor derivatives were synthesized via microwave Buchwald-Hartwig cross-coupling reactions with yields of up to 91%. The compounds exhibit three different types of photoluminescence, which is well-explained by quantum chemical calculations.

Binary AsSe alloys from the border of a glass-forming region (65 < < 70) subjected to nanomilling in dry and dry-wet modes are characterized by the XRPD, micro-Raman scattering (micro-RS) and revised positron annihilation lifetime (PAL) methods complemented by a disproportionality analysis using the quantum-chemical cluster modeling approach. These alloys are examined with respect to tetra-arsenic biselenide AsSe stoichiometry, realized in glassy g-AsSe, glassy-crystalline g/c-AsSe and glassy-crystalline g/c-AsSe. From the XRPD results, the number of rhombohedral As and cubic arsenolite AsO phases in As-Se alloys increases after nanomilling, especially in the wet mode realized in a PVP water solution.

Polyamorphic transformations driven by high-energy mechanical ball milling (nanomilling) are recognized in a melt-quenched glassy alloy of tetra-arsenic triselenide (AsSe). We employed XRPD analysis complemented by thermophysical heat-transfer and micro-Raman spectroscopy studies. A straightforward interpretation of the medium-range structural response to milling-driven reamorphization is developed within a modified microcrystalline model by treating diffuse peak-halos in the XRPD patterns of this alloy as a superposition of the Bragg-diffraction contribution from inter-planar correlations, which are supplemented by the Ehrenfest-diffraction contribution from inter-atomic and/or inter-molecular correlations related to derivatives of thioarsenide AsSe molecules, mainly dimorphite-type AsSe ones.
