Ecological Profile of the Flea Beetle Genus Heyden in South Africa (Chrysomelidae, Galerucinae, Alticini).

In this work, the 25 species of the flea beetle genus Heyden recorded for South Africa are considered. Starting from the updated species distribution and the topographic, temperature, and precipitation variables, as well as the vegetation types in the occurrence sites, through an analysis of ecological niche modelling, a possible ecological profile is provided, both for each species and the entire genus, highlighting some of the factors that drive their occurrence and distribution patterns. Along with the vegetation type, some climatic variables were found to be particularly influential, such as the mean temperature of both the wettest and driest quarters and also the mean precipitation of the wettest period.

We report the design and development of a novel multifunctional nanostructure, RB-AuSiO_HSA-DOX, where tri-modal cancer treatment strategies-photothermal therapy (PTT), photodynamic therapy (PDT), chemotherapy-luminescent properties and targeting are integrated into the same scaffold. It consists of a gold core with optical and thermo-plasmonic properties and is covered by a silica shell entrapping a well-known photosensitizer and luminophore, Rose Bengal (RB). The nanoparticle surface was decorated with Human Serum Albumin (HSA) through a covalent conjugation to confer its targeting abilities and as a carrier of Doxorubicin (DOX), one of the most effective anticancer drugs in clinical chemotherapy.

Liquid biopsy (LB) involves the analysis of circulating tumour-derived DNA (ctDNA), providing a minimally invasive method for gathering both quantitative and qualitative information. Genomic analysis of ctDNA through next-generation sequencing (NGS) enables comprehensive genetic profiling of tumours, including non-driver alterations that offer prognostic insights. LB can be applied in both early-stage disease settings, for the diagnosis and monitoring of minimal residual disease (MRD), and advanced disease settings, for monitoring treatment response and understanding the mechanisms behind disease progression and tumour heterogeneity.

This study examined the response to cisplatin in BxPC-3, Mia-Paca-2, PANC-1, and YAPC pancreatic cancer lines with different genotypic and phenotypic characteristics, and the mechanisms associated with their resistance. BxPC-3 and MIA-PaCa-2 cell lines were the most sensitive to cisplatin, while YAPC and PANC-1 were more resistant. Consistently, in cisplatin-treated BxPC-3 cells, the cleavage patterns of pro-caspase-9, -7, -3, and PARP-1 demonstrated that they were more sensitive than YAPC cells.

Increased expression of branched-chain amino acid (BCAA) transaminase 1 (BCAT1) often correlates with tumor aggressiveness and drug resistance in cancer. We have recently reported that BCAT1 was overexpressed in a subgroup of T-cell acute lymphoblastic (T-ALL) samples, especially those with NOTCH1 activating mutations. Interestingly, BCAT1-depleted cells showed pronounced sensitivity to DNA-damaging agents such as etoposide; however, how BCAT1 regulates this sensitivity remains uncertain.
