Ravulizumab is a second-generation complement component 5 (C5) inhibitor (C5i) approved for the treatment of paroxysmal nocturnal hemoglobinuria (PNH) following positive results from two pivotal trials in patients with PNH originally naive to C5i treatment and eculizumab-experienced patients with PNH. In both trials, after the 26week primary evaluation period, all patients received ravulizumab for up to 6 years. To report ravulizumab treatment outcomes in patients with PNH originally naive to C5i treatment and eculizumab-experienced patients with PNH treated for up to 6 years.

Nutritional immunity, a key component of the vertebrate innate immune response, involves the modulation of zinc availability to limit the growth of pathogens. counteracts host-imposed zinc starvation through metabolic adaptations, including reprogramming of gene expression and activating efficient metal uptake systems. Pancreatic ductal adenocarcinoma (PDAC) contains an extensive stroma that modulates response to therapy, contributing to the dismal prognosis associated with this cancer. Evidence suggests that PDAC stromal composition is shaped by mutations within malignant cells, but most previous work has focused on pre-clinical models driven by KrasG12D and mutant Trp53. Elucidation of the contribution of additional known oncogenic drivers, including KrasG12V mutation and Smad4 loss, is needed to increase understanding of malignant cell-stroma crosstalk in PDAC.

Developmental Language Disorder (DLD) is a clinical condition characterised by language difficulties without cognitive or neurological impairments, leading to communication and learning challenges. The study explores the narrative and linguistic abilities of children with DLD and Typically Developing (TD) peers by analysing both macrostructural and microstructural aspects of their narrative production elicited during a storytelling task and describing the types of grammatical and lexical errors. Participants included 19 children with expressive DLD aged 4-8 years and 19 TD children matched by age and gender.

Purpose To assess the impact of scanner manufacturer and scan protocol on the performance of deep learning models to classify prostate cancer (PCa) aggressiveness on biparametric MRI (bpMRI). Materials and Methods In this retrospective study, 5,478 cases from ProstateNet, a PCa bpMRI dataset with examinations from 13 centers, were used to develop five deep learning (DL) models to predict PCa aggressiveness with minimal lesion information and test how using data from different subgroups-scanner manufacturers and endorectal coil (ERC) use (Siemens, Philips, GE with and without ERC and the full dataset)-impacts model performance. Performance was assessed using the area under the receiver operating characteristic curve (AUC).

PADI4 is one of the human isoforms of a family of enzymes involved in the conversion of arginine to citrulline. MDM2 is an E3 ubiquitin ligase that is critical for degradation of the tumor suppressor gene p53. We have previously shown that there is an interaction between MDM2 and PADI4 in cellulo, and that such interaction occurs through the N-terminal region of MDM2, N-MDM2, and in particular through residues Thr26, Val28, Phe91, and Lys98.

A multiscale quantum mechanical (QM)/classical approach is presented that is able to model the optical properties of complex nanostructures composed of a molecular system adsorbed on metal nanoparticles. The latter is described by a combined atomistic-continuum model, where the core is described using the implicit boundary element method (BEM) and the surface retains a fully atomistic picture and is treated employing the frequency-dependent fluctuating charge and fluctuating dipole (ωFQFμ) approach. The integrated QM/ωFQFμ-BEM model is numerically compared with state-of-the-art fully atomistic approaches, and the quality of the continuum/core partition is evaluated.
