Janus kinase (JAK) inhibitors provide limited depth and durability of response in myelofibrosis. We evaluated pelabresib-a bromodomain and extraterminal domain (BET) inhibitor-plus ruxolitinib (a JAK inhibitor) compared with placebo plus ruxolitinib as first-line therapy. In this phase 3 study (MANIFEST-2), JAK inhibitor-naive patients with myelofibrosis were randomized 1:1 to pelabresib 125 mg once daily (QD; 50-175 mg QD permitted) for 14 days followed by a 7-day break (21-day cycle), or to placebo in combination with ruxolitinib 10 or 15 mg twice daily (BID; 5 mg QD-25 mg BID permitted).

Infants born 22-23 weeks have more immature organ development and physiology than more mature extremely low birth weight infants. Objectives: This retrospective cohort study aimed to assess the long-term growth and associated risk factors of macular atrophy (MA) in eyes with neovascular age-related macular degeneration (nAMD) treated with intravitreal anti-vascular endothelial growth factor (anti-VEGF) compounds.
Methods: Two hundred and six patients initiating anti-VEGF therapy were followed for 8 years using a treat-and-extend protocol. The study analysed correlations between MA growth (by square root transformation measured in infrared images) and clinical parameters, and genetic variants for AMD in the complement and lipid pathways and the ARMS2 gene.

Objective: To estimate the prevalence of true knot of the umbilical cord (TKUC) and identify associated adverse maternal and neonatal outcomes.
Study Design: A multicenter retrospective cohort study was conducted, including all women with singleton pregnancies who delivered between 24 and 42 weeks of gestation from 2005 to 2021 at two large obstetrical centers. Gross pathological examinations of the placenta and umbilical cord were routinely performed immediately after delivery.

Background: Respiratory syncytial virus (RSV) is a major cause of severe respiratory infections in infants, especially under 5 years of age. Several studies have reported that interactions between RSV and bacteria like Streptococcus pneumoniae (SP), Moraxella catarrhalis (MC), Haemophilus influenzae (HI) and Staphylococcus aureus (SA) may influence their pathogenicity and the clinical outcome of infection. Morally injurious events (MIEs), encompassing personal transgressions, witnessing others commit transgressions, or experiencing betrayal by leaders, can conflict with one's moral/ethical principles, evoking outrage and profound mistrust. Although MIEs are associated with depression, PTSD, and anxiety, the mechanisms linking MIEs to psychiatric symptomatology remain unclear, especially among civilians in times of collective trauma. This study explored one potential mechanism: stigma toward mental-health treatment, which can deter help-seeking and exacerbate guilt, shame, and mistrust.
