Controlling the size of powder particles is pivotal in the design of many pharmaceutical forms and the related manufacturing processes and plants. One of the most common techniques for particle size reduction in the process industry is powder milling, whose efficiency relates to the mechanical properties of the powder particles themselves. In this work, we first characterize the elastic and plastic responses of different pharmaceutical powders by measuring their Young modulus, the hardness, and the brittleness index via nano-indentation.

Single-cell multi-omics assays offer unprecedented opportunities to explore epigenetic regulation at cellular level. However, high levels of technical noise and data sparsity frequently lead to a lack of statistical power in correlative analyses, identifying very few, if any, significant associations between different molecular layers. Here we propose SCRaPL, a novel computational tool that increases power by carefully modelling noise in the experimental systems.

Ultracold atoms confined to periodic potentials have proven to be a powerful tool for quantum simulation of complex many-body systems. We confine fermions to one dimension to realize the Tomonaga-Luttinger liquid model, which describes the highly collective nature of their low-energy excitations. We use Bragg spectroscopy to directly excite either the spin or charge waves for various strengths of repulsive interaction.

A common approach for studying a solid solution or disordered system within a periodic framework is to create a supercell in which certain amounts of target elements are substituted with other elements. The key to generating supercells is determining how to eliminate symmetry-equivalent structures from many substitution patterns. Although the total number of substitutions is on the order of trillions, only symmetry-inequivalent atomic substitution patterns need to be identified, and their number is far smaller than the total.

The present review focuses on the physiological states of spinal networks, which are stochastically modulated by continuously changing ensembles of proprioceptive and supraspinal input resulting in highly redundant neural networks. Spinal epidural interfaces provide a platform for probing spinal network dynamics and connectivity among multiple motor pool-specific spinal networks post-injury under in vivo experimental conditions. Continuous epidural low-frequency pulses at low intensity can evoke motor responses of stochastically changing amplitudes and with an oscillatory pattern of modulation.

Serial dependence entails an attractive bias based on the recent history of stimulation, making the current stimulus appear more similar to the preceding one. Although serial dependence is ubiquitous in perception, its nature and mechanisms remain unclear. Here, in two independent experiments, we test the hypothesis that this bias originates from high-level processing stages at the level of abstract information processing (Exp.

The effect of atomic relaxations on the temperature-dependent elastic constants (TDECs) is usually taken into account at zero temperature by the minimization of the total energy at each strain. In this paper, we investigate the order of magnitude of this approximation on a paradigmatic example: the C elastic constant of diamond and zincblende materials. We estimate the effect of finite-temperature atomic relaxations within the quasi-harmonic approximation by computing ab initio the internal strain tensor from the second derivatives of the Helmholtz free-energy with respect to strain and atomic displacements.

Emerging evidence suggests a role of the posterior cortices in regulating alpha oscillatory activity and organizing low-level processing in non-alpha frequency bands. Therefore, posterior brain lesions, which damage the neural circuits of the visual system, might affect functional connectivity patterns of brain rhythms. To test this hypothesis, eyes-closed resting state EEG signal was acquired from patients with hemianopia with left and right posterior lesions, patients without hemianopia with more anterior lesions and healthy controls.

Epitopes that bind simultaneously to all human alleles of Major Histocompatibility Complex class II (MHC II) are considered one of the key factors for the development of improved vaccines and cancer immunotherapies. To engineer MHC II multiple-allele binders, we developed a protocol called PanMHC-PARCE, based on the unsupervised optimization of the epitope sequence by single-point mutations, parallel explicit-solvent molecular dynamics simulations and scoring of the MHC II-epitope complexes. The key idea is accepting mutations that not only improve the affinity but also reduce the affinity gap between the alleles.

The possible neurodevelopmental consequences of SARS-CoV-2 infection are presently unknown. In utero exposure to SARS-CoV-2 has been hypothesized to affect the developing brain, possibly disrupting neurodevelopment of children. Spike protein interactors, such as ACE2, have been found expressed in the fetal brain, and could play a role in potential SARS-CoV-2 fetal brain pathogenesis.

Mechanical stiffness, as opposed to softness, is a fundamental property of solids. Its persistence or rheological evolution in vibrating solid-solid nanocontacts is important in physics, materials science and technology. A puzzling apparent liquefaction under oscillatory strain, totally unexpected at room temperature, was suggested by recent experiments on solid gold nano-junctions.

High Mobility Group A1 (HMGA1) is an architectural chromatin factor involved in the regulation of gene expression and a master regulator in Triple Negative Breast Cancer (TNBC). In TNBC, HMGA1 is overexpressed and coordinates a gene network that controls cellular processes involved in tumour development, progression, and metastasis formation. Here, we find that the expression of HMGA1 and of the microtubule-destabilizing protein stathmin correlates in breast cancer (BC) patients.

The peeling of an immobile adsorbed membrane is a well-known problem in engineering and macroscopic tribology. In the classic setup, picking up at one extreme and pulling off result in a peeling force that is a decreasing function of the pickup angle. As one end is lifted, the detachment front retracts to meet the immobile tail.

Background: The COVID-19 outbreak imposed an overwhelming workload as well as emotional burdens on Healthcare workers (HCWs). In May 2020, an online survey was administered to HCWs in Italy to assess the pandemic's psychological impact and to investigate possible predictive factors that led to individual differences.

Methods: The psychological experience was measured based on the prevalence of self-reported feelings during the pandemic, including negative and positive emotional states.

In periodic systems of interacting electrons, Fermi and Luttinger surfaces refer to the locations within the Brillouin zone of poles and zeros, respectively, of the single-particle Green's function at zero energy and temperature. Such difference in analytic properties underlies the emergence of well-defined quasiparticles close to a Fermi surface, in contrast to their supposed non-existence close to a Luttinger surface, where the single-particle density-of-states vanishes at zero energy. We here show that, contrary to such common belief, dispersive 'quasiparticles' with infinite lifetime do exist also close to a pseudo-gapped Luttinger surface.

Microbial plant pathogens secrete a range of effector proteins that damage host plants and consequently constrain global food production. Necrosis and ethylene-inducing peptide 1-like proteins (NLPs) are produced by numerous phytopathogenic microbes that cause important crop diseases. Many NLPs are cytolytic, causing cell death and tissue necrosis by disrupting the plant plasma membrane.

Towards SINEUP-based therapeutics: Design of an synthesized SINEUP RNA.

Mol Ther Nucleic Acids

March 2022

Central RNA Laboratory, Istituto Italiano di Tecnologia (IIT), 16152 Genova, Italy.

SINEUPs are a novel class of natural and synthetic non-coding antisense RNA molecules able to increase the translation of a target mRNA. They present a modular organization comprising an unstructured antisense target-specific domain, which sets the specificity of each individual SINEUP, and a structured effector domain, which is responsible for the translation enhancement. In order to design a fully functional transcribed SINEUP for therapeutics applications, SINEUP RNAs were synthesized with a variety of chemical modifications and screened for their activity on endogenous target mRNA upon transfection.

Deprivation of Auditory Experience Influences Numerosity Discrimination, but Not Numerosity Estimation.

Brain Sci

January 2022

U-VIP, Unit for Visually Impaired People, Istituto Italiano di Tecnologia, 16163 Genova, Italy.

Number sense is the ability to estimate the number of items, and it is common to many species. Despite the numerous studies dedicated to unveiling how numerosity is processed in the human brain, to date, it is not clear whether the representation of numerosity is supported by a single general mechanism or by multiple mechanisms. Since it is known that deafness entails a selective impairment in the processing of temporal information, we assessed the approximate numerical abilities of deaf individuals to disentangle these two hypotheses.

Article Synopsis
  • There is evidence that the mechanical properties of central nervous system (CNS) neurons influence cellular activities, particularly through mechanical stimuli.
  • Using oscillatory optical tweezers, researchers found that a local indentation force greater than 13 pN triggers a brief increase in intracellular calcium levels, while repeated stimulations lead to a sustained response.
  • The study indicates that these mechanical signals rely on calcium entering through ion channels, which activates specific proteins like CAMKII and RhoA, highlighting the importance of mechanical signaling in neuronal networks.
