Electronic address:

Article Synopsis
  • Patients with spontaneous coronary artery dissection (SCAD) who presented as ST-segment-elevation myocardial infarction (STEMI) exhibited more severe coronary lesions and worse blood flow compared to those presenting as non-ST-segment elevation myocardial infarction (NSTEMI).
  • During treatment, STEMI patients were more frequently treated with percutaneous coronary intervention (PCI) and had higher rates of left ventricular systolic dysfunction, but both groups had similar rates of major adverse events during hospital admission.
  • In the long term, despite the initial differences in severity and treatment, both STEMI and NSTEMI SCAD patients had comparable outcomes concerning major adverse cardiac and cerebrovascular events.

Pre-blockade of the sarco-endoplasmic reticulum (ER) calcium ATPase (SERCA) with irreversible thapsigargin depresses exocytosis in adrenal bovine chromaffin cells (BCCs). Distinct expression of voltage-dependent Ca-channel subtypes and of the Ca-induced Ca release (CICR) mechanism in BCCs versus mouse chromaffin cells (MCCs) has been described. We present a parallel study on the effects of the acute SERCA blockade with reversible cyclopizonic acid (CPA), to repeated pulsing with acetylcholine (ACh) at short (15 s) and long intervals (60 s) at 37 °C, allowing the monitoring of the initial size of a ready-release vesicle pool (RRP) and its depletion and recovery in subsequent stimuli.

The SIN3A histone deacetylase complex is required for a complete transcriptional response to hypoxia.

Nucleic Acids Res

January 2018

Departamento de Bioquímica, Universidad Autónoma de Madrid (UAM) and Instituto de Investigaciones Biomédicas 'Alberto Sols' (CSIC-UAM), 28029 Madrid, Spain.

Cells adapt to environmental changes, including fluctuations in oxygen levels, through the induction of specific gene expression programs. To identify genes regulated by hypoxia at the transcriptional level, we pulse-labeled HUVEC cells with 4-thiouridine and sequenced nascent transcripts. Then, we searched genome-wide binding profiles from the ENCODE project for factors that correlated with changes in transcription and identified binding of several components of the Sin3A co-repressor complex, including SIN3A, SAP30 and HDAC1/2, proximal to genes repressed by hypoxia.

Identification of non-coding genetic variants in samples from hypoxemic respiratory disease patients that affect the transcriptional response to hypoxia.

Nucleic Acids Res

November 2016

Departamento de Bioquímica, Universidad Autónoma de Madrid (UAM) and Instituto de Investigaciones Biomédicas 'Alberto Sols' (CSIC-UAM), 28029 Madrid, Spain

A wide range of diseases course with an unbalance between the consumption of oxygen by tissues and its supply. This situation triggers a transcriptional response, mediated by the hypoxia inducible factors (HIFs), that aims to restore oxygen homeostasis. Little is known about the inter-individual variation in this response and its role in the progression of disease.
