The growth of microbial multidrug resistance is a problem in modern clinical medicine. Chemical modification of active pharmaceutical ingredients is an attractive strategy to improve their biopharmaceutical properties by increasing bioavailability and reducing drug toxicity. We present results of MD and MC simulations of the equilibrium properties of swelling gels with comb-like or bottlebrush subchains and compare them to scaling-theory predictions. In accordance with theory, the simulation results demonstrate that swelling coefficient of the gel increases as a function of the polymerization degree of the main chains and exhibits a very weak maximum (or is virtually constant) as a function of the polymerization degree and grafting density of side chains. The bulk osmotic modulus passes through a shallow minimum as the polymerization degree of the side chains increases.

In the present work, we address the problem of utilizing machine learning (ML) methods to predict the thermal properties of polymers by establishing "structure-property" relationships. Having focused on a particular class of heterocyclic polymers, namely polyimides (PIs), we developed a graph convolutional neural network (GCNN), being one of the most promising tools for working with big data, to predict the PI glass transition temperature as an example of the fundamental property of polymers. To train the GCNN, we propose an original methodology based on using a "transfer learning" approach with an enormous "synthetic" data set for pretraining and a small experimental data set for its fine-tuning.

Regular and irregular molecular brushes with polydimethylsiloxane backbone and poly-2-isopropyl-2-oxazoline side chains have been synthesized. Prepared samples differed strongly in the side chain grafting density, namely, in the ratio of the lengths of spacer between the grafting points and the side chains. The hydrodynamic properties and molecular conformation of the synthesized grafted copolymers and their behavior in aqueous solutions on heating were studied by the methods of molecular hydrodynamics and optics.

Mucoadhesive polymer patches are a promising alternative for prolonged and controlled delivery of topical corticosteroids (CS) to improve their biopharmaceutical properties (mainly increasing local bioavailability and reducing systemic toxicity). The main biopharmaceutical advantages of patches compared to traditional oral dosage forms are their excellent bioadhesive properties and their increased drug residence time, modified and unidirectional drug release, improved local bioavailability and safety profile, additional pain receptor protection, and patient friendliness. This review describes the main approaches that can be used for the pharmaceutical R&D of oromucosal patches with improved physicochemical, mechanical, and pharmacological properties.

H spin-diffusion solid-state NMR was utilized to elucidate the domain size in multiblock-copolymers (BCPs) poly-( poly(dimethylsiloxane)- ladder-like poly(phenylsiloxane)) and poly-( poly((3,3',3″-trifluoropropyl-methyl)siloxane)- ladder-like poly(phenylsiloxane). It was found that these BCPs form worm-like morphology with rigid cylinders dispersed in amorphous matrix. By using the combination of solid-state NMR techniques such as C CP/MAS, C direct-polarization MAS and 2D H EXSY, it was shown that the main factor which governs the diameter value of these rigid domains is the presence of interpenetrating segments of soft blocks.

Polyelectrolyte complexes (PECs), based on partially deacetylated chitin nanowhiskers (CNWs) and anionic polysaccharides, are characterized by their variability of properties (particle size, ζ-potential, and pH-sensitivity) depending on the preparation conditions, thereby allowing the development of polymeric nanoplatforms with a sustained release profile for active pharmaceutical substances. This study is focused on the development of hydrogels based on PECs of CNWs and sodium alginate (ALG) for potential vaginal administration that provide controlled pH-dependent antibiotic release in an acidic vaginal environment, as well as prolonged pharmacological action due to both the sustained drug release profile and the mucoadhesive properties of the polysaccharides. The desired hydrogels were formed as a result of both electrostatic interactions between CNWs and ALG (PEC formation), and the subsequent molecular entanglement of ALG chains, and the formation of additional hydrogen bonds.

The interaction of colloidal particles with a planar polymer brush immersed in a solvent of variable thermodynamic quality is studied by a numerical self-consistent field method combined with analytical mean-field theory. The effect of embedded particle on the distribution of polymer density in the brush is analyzed and the particle insertion free energy profiles are calculated for variable size and shape of the particles and sets of polymer-particle and polymer-solvent interaction parameters. In particular, both cases of repulsive and attractive interactions between particles and brush-forming chains are considered.

Swelling behaviour and bulk moduli of polymer gels comprising of crosslinked bottlebrush subchains enable fine tuning by varying polymerization degrees of the main and side chains of the bottlebrush strands as well as their grafting densities. By using scaling approach we predict power law dependences of structural and elastic properties of swollen bottlebrush gels on the set of relevant architectural parameters and construct phase diagrams consisting of regions corresponding to different power law asymptotics for these dependences. In particular, our theory predict that bulk elastic modulus of the gel exhibits non-monotonous dependence on the degree of polymerization of side chains of the bottlebrush strands.

To study conformational transition occuring upon inferior solvent strength in a brush formed by linear or dendritically branched macromolecules tethered to the inner surface of cylindrical or planar (slit-like) pore, a self-consistent field analytical approach is employed. Variations in the internal brush structure as a function of variable solvent strength and pore radius, and the onset of formation of a hollow channel in the pore center are analysed. We present a scaling theory describing the equilibrium properties of spherical micelles formed by a diblock copolymer with bottlebrush blocks in a selective solvent. The theory predicts a number of new thermodynamic regimes inherent for copolymers with relatively short main chains (long side chains) in the bottlebrush blocks. These regimes with a novel set of scaling exponents for the micelle properties are characterized by limiting extension of the main chains of the core or/and corona-forming blocks and do not exist for micelles of conventional linear block copolymers.

The development of intravitreal glucocorticoid delivery systems is a current global challenge for the treatment of inflammatory diseases of the posterior segment of the eye. The main advantages of these systems are that they can overcome anatomical and physiological ophthalmic barriers and increase local bioavailability while prolonging and controlling drug release over several months to improve the safety and effectiveness of glucocorticoid therapy. One approach to the development of optimal delivery systems for intravitreal injections is the conjugation of low-molecular-weight drugs with natural polymers to prevent their rapid elimination and provide targeted and controlled release.
