Genetic association between mitochondrial DNA copy number and glioma risk: insights from causality.

BMC Cancer

November 2024

Department of Neurosurgery, West China Hospital, Sichuan University, 37 Guoxue Lane, Wuhou District, Chengdu, 610041, Sichuan, China.

Article Synopsis
  • The study investigated the link between mitochondrial DNA copy number (mtDNA-CN) and the development of glioma and glioblastoma (GBM) using data from large health datasets.
  • The analysis found no significant causal relationship between mtDNA-CN and glioma or low-grade glioma, while a potential association with GBM was suggested.
  • The results were confirmed through various robust statistical analyses, leading to the conclusion that while mtDNA-CN does not impact glioma risk, it may play a role in the development of GBM.

Interlaboratory consistency of promoter methylation detection in colorectal cancer using the post-optimized materials.

iScience

November 2024

National Center for Clinical Laboratories, Institute of Geriatric Medicine, Chinese Academy of Medical Sciences, Beijing Hospital/ National Center of Gerontology, Beijing, P.R. China.

Fecal DNA-based Syndecan 2 () methylation detection is a promising non-invasive strategy for early colorectal cancer (CRC) screening. In China, commercial assays for methylation detection vary in sensitivity and specificity, yet there is no standardized external quality assessment (EQA) to ensure accuracy. This study utilized CRISPR-Cas9 and homology-directed repair (HDR) technologies to edit the promoter in 293T cells, creating hypermethylated and heterogeneous cell lines.

Association between homocysteine levels and mortality in CVD: a cohort study based on NHANES database.

BMC Cardiovasc Disord

November 2024

Department of Cardiology, Beijing Hospital, National Center of Gerontology; Institute of Geriatric Medicine, Chinese Academy of Medical Sciences, Beijing, People's Republic of China.

Background: Cardiovascular disease (CVD) is a major global health concern with increasing incident cases and deaths. Homocysteine (Hcy) has been investigated for its potential association with CVD, researchers have debated the extent to which Hcy should be considered a risk factor for cardiovascular diseases, as only 50% of CVD can be explained by classical risk factors.

Methods: We conducted a prospective cohort study using NHANES 1999-2006 data, analyzing 1,739 US patients aged at least 30 with CVD.

Objective: There is controversy among different guidelines regarding the use of thermal ablation to treat clinical T1a renal cell carcinomas with tumor sizes ranging from 3.1-4 cm. Therefore, we compared oncological outcomes between heat-based thermal ablation (hTA) and cryoablation (CA) in patients with solid T1a renal cell carcinomas, including those with a tumor size ≤3 cm and a tumor size of 3.

Relapse is one of the major challenges in clinical treatment of acute myeloid leukemia (AML). Though minimal residual disease (MRD) monitoring plays a crucial role in quantitative assessment of the disease, molecular MRD analysis has been mainly limited to patients diagnosed with gene fusions and NPM1 mutations. Here, we report a longitudinal ultra-sensitive mutation burden (UMB) monitoring strategy for accurate MRD analysis in AML patients regardless of genetic abnormality types.

Article Synopsis
  • - Recent research indicates that ribociclib, a CDK4/6 inhibitor, is linked to serious heart-related side effects, particularly QT prolongation, although studies show mixed results on cardiotoxic impact across different inhibitors.
  • - Using data from the FDA adverse event reporting system, the study found considerable variation in heart issues among CDK4/6 inhibitors, with ribociclib showing the highest risk, particularly for severe cardiotoxic events like torsades de pointes.
  • - Factors like age, location, and other medications significantly affect these risks, highlighting the importance of tailored treatment plans to reduce cardiotoxicity in breast cancer patients.

Evaluation and prediction of relapse risk in stable systemic lupus erythematosus patients after glucocorticoid withdrawal (PRESS): an open-label, multicentre, non-inferiority, randomised controlled study in China.

Ann Rheum Dis

November 2024

Department of Rheumatology, Beijing Hospital, National Centre of Gerontology, Institute of Geriatric Medicine, Clinical Immunology Centre, Chinese Academy of Medical Sciences, Beijing, China

Article Synopsis
  • The study aimed to analyze the relapse rates in systemic lupus erythematosus (SLE) patients after stopping glucocorticoids (GC) with or without hydroxychloroquine (HCQ) maintenance therapy.
  • Conducted over 33 weeks, the trial divided 333 patients into three groups: one that stopped both drugs, one that only stopped GC while continuing HCQ, and one that continued both treatments.
  • Results showed higher relapse rates in the drug-free group (26.1%) compared to both HCQ (11.2%) and dual maintenance (4.7%) groups, indicating that continuing HCQ could help prevent relapses after stopping GC.

Pulmonary embolism (PE) is one of the leading causes of cardiovascular death, resulting in a significant socioeconomic burden. Although current treatments primarily focus on anticoagulation and thrombolysis, there is an urgent need for a better understanding of its pathophysiology and the development of new treatment strategies. Animal models play a crucial role in understanding PE and developing new therapies for the disease, with rodents commonly used due to ethical and cost considerations.

Objective: Blood-stasis syndrome (BSS), an important syndrome in Type 2 diabetes mellitus(T2DM), is associated with the pathophysiological mechanisms underlying diabetic vascular complications. However, BSS has not been fully characterized as of yet. Due to the strong correlation between BSS and vasculopathy, we hypothesized that the metabolic characteristics of BSS in T2DM (T2DM BSS) are highly specific.
