Expressmarker mtDNA-SNP60 kit, a 3-dye fluorescence labeling forensic mitochondrial-SNP kit, was developed and modified based on previous work. In this study, it was validated through sensitivity, precision, accuracy, reproducibility, species specificity, mixture samples, PCR-based conditions, and inhibitor studies, according to the Scientific Working Group on DNA Analysis Methods guidelines. In the sensitivity study, full profile was obtained from template DNA with the quantity ≥ 4 pg.

Investigations into the use of mitochondrial genome (mtGenome) typing by massively parallel sequencing technologies are well underway in many areas, including forensic genetics. Previous studies have demonstrated that mtGenome sequencing data generated from Ion torrent personal genome machine (PGM) system were highly viable and reliable in forensic research. In recent years, forensic scientists have focused on the discrimination of body fluids using microbial signatures. In this study, we performed PCR-based detection of microbial signatures of vaginal fluid, saliva, and feces in a Han Chinese population. We investigated the 16S rRNA genes of Lactobacillus crispatus, Lactobacillus gasseri, Lactobacillus jensenii, Lactobacillus iners, and Atopobium vaginae in vaginal fluid, the 16S rRNA and the glucosyltransferase enzyme genes of Streptococcus salivarius and Streptococcus mutans in saliva, and the 16S rRNA genes of Enterococcus species, the RNA polymerase β-subunit gene of Bacteroides uniformis and Bacteroides vulgatus, and the α-1-6 mannanase gene of Bacteroides thetaiotaomicron in feces.

Zeolitic imidazolate frameworks (ZIFs) consist of metal nodes connected to imidazolate linkers, having both the properties of metal-organic frameworks (MOFs) and inorganic zeolites, such as controllable pore sizes, high porosity and surface areas, as well as exceptional thermal and chemical stability, thereby making them a class of attractive materials for diverse analytical applications. In this study, we reported a facile magnetization process of ZIF-7 (zinc benzimidazolate) for simultaneous magnetic extraction of polycyclic aromatic hydrocarbons (PAHs) by simply mixing ZIF-7 and polydopamine (PDA)-coated Fe3O4 nanoparticles (PDA@Fe3O4) in solutions. Functional groups (-OH and -NH2), provided by PDA as a highly efficient molecular linker, could attract and anchor ZIF-7 through noncovalent adsorption and covalent cross-link interactions, thereby promoting the complete magnetization of ZIFs and enhancing their stability and reusability.

As our ongoing work on research of gelatinase inhibitors, an array of hydrazide-containing peptidomimetic derivatives bearing quinoxalinone as well as spiro-heterocyclic backbones were designed, synthesized, and assayed for their in vitro enzymatic inhibitory effects. The results demonstrated that both the quinoxalinone (series I and II) and 1,4-dithia-7-azaspiro[4,4]nonane-based hydrazide peptidomimetics (series III) displayed remarkably selectivity towards gelatinase A as compared to APN, with IC50 values in the micromole range. Structure-activity relationships were herein briefly discussed.

The low mutation rates, short amplicon sizes and length variation characteristics of biallelic insertion/deletion polymorphisms (INDELs) are features that allow combining advantages of both STR and SNP markers for genetic analysis. The Qiagen Investigator DIPplex kit contains 30 biallelic autosomal INDELs plus Amelogenin for forensic personal identification and parentage testing. In this work, an ordered mesoporous carbon (OMC)/Nafion coated fiber for solid-phase microextraction (SPME) was prepared and used as the working electrode for electro-enhanced SPME (EE-SPME) of amphetamines. The EE-SPME strategy is primarily based on the electro-migration and complementary charge interaction between fiber coating and ionic compounds. Compared with traditional SPME, EE-SPME exhibited excellent extraction efficiency for amphetamine (AP) and methamphetamine (MA) with an enhancement factor of 7.
