Type II topoisomerases shape multi-scale 3D chromatin folding in regions of positive supercoils.

Mol Cell

November 2024

Institute of Molecular Biology gGmbH, Ackermannweg 4, 55128 Mainz, Germany; Department of General Biology, Medical School, University of Patras, Rio, Patras 26500 Greece. Electronic address:

Type II topoisomerases (TOP2s) resolve torsional stress accumulated during various cellular processes and are enriched at chromatin loop anchors and topologically associated domain (TAD) boundaries, where, when trapped, can lead to genomic instability promoting the formation of oncogenic fusions. Whether TOP2s relieve topological constraints at these positions and/or participate in 3D chromosome folding remains unclear. Here, we combine 3D genomics, imaging, and GapRUN, a method for the genome-wide profiling of positive supercoiling, to assess the role of TOP2s in shaping chromosome organization in human cells.

Lung nodule localization and size estimation on chest tomosynthesis.

J Med Imaging (Bellingham)

January 2025

University of Gothenburg, Sahlgrenska Academy, Institute of Clinical Sciences, Department of Radiology, Gothenburg, Sweden.

Purpose: We aim to investigate the localization, visibility, and measurement of lung nodules in digital chest tomosynthesis (DTS).

Approach: Computed tomography (CT), maximum intensity projections (CT-MIP) (transaxial versus coronal orientation), and computer-aided detection (CAD) were used as location reference, and inter- and intra-observer agreement regarding lung nodule size was assessed. Five radiologists analyzed DTS and CT images from 24 participants with lung , focusing on lung nodule localization, visibility, and measurement on DTS.

Maternal diet in pregnancy and the risk of inflammatory bowel disease in the offspring: a prospective cohort study.

Am J Clin Nutr

January 2025

Pediatric Research Institute, Oslo University Hospital and Faculty of Medicine, University of Oslo, Oslo, Norway; Department of Child and Adolescent Health, Oslo University Hospital, Oslo, Norway.

Article Synopsis
  • This study investigates the link between maternal diet during pregnancy and the risk of inflammatory bowel disease (IBD) in children, using data from over 85,000 Norwegian children born between 1999 and 2009.
  • Results show that higher maternal diet diversity during pregnancy significantly lowers the risk of ulcerative colitis (UC) in children, while findings for Crohn's disease (CD) and overall IBD were not as strong.
  • The research suggests that improving diet diversity for pregnant mothers might help reduce the risk of UC in their children, highlighting the importance of maternal nutrition.

Article Synopsis
  • The study compares the cost-effectiveness of cemented versus uncemented hip hemiarthroplasty (HHA) for elderly patients with femoral neck fractures, using a Markov model and Monte Carlo simulation for analysis.
  • Results showed that while the upfront costs of cemented HHA are higher, it had lower total costs overall ($19,462 vs. $21,997) and yielded more quality-adjusted life years (4.0 vs. 3.1).
  • The conclusion indicates that cemented HHA is the more cost-effective option, being the preferred treatment for 89% of patients in the simulation, despite higher initial expenses.
