Objective: The blood pressure (BP) response to salt intake (salt sensitivity) shows great variability among individuals and is more frequent in hypertensive patients. Elevated levels of the steroid hormone Endogenous Ouabain (EO) are associated with hypertension (HT) and salt sensitivity. The lanosterol synthase gene ( LSS ) plays a key role in the biosynthesis of steroids and its rs2254524 variant (Val642Leu) is linked to salt sensitivity in humans.

Evaluation of treatment outcomes in patients supported by temporary mechanical circulatory support (tMCS) currently relies mainly on mortality, which may not sufficiently address other patient benefits or harms. Bleeding and thrombosis are major contributors to mortality. Still, current bleeding scores are not designed for critically ill patients undergoing tMCS, only consider selected populations, and do not account for the high heterogeneity among bleeding and thrombotic adverse events.

Background: Stroke prevention with direct-acting oral anticoagulant agents in patients with atrial fibrillation confers a risk of bleeding and limits their use. Asundexian, an activated factor XI (XIa) inhibitor, is an oral anticoagulant that may prevent strokes with less bleeding.

Methods: In a phase 3, international, double-blind trial, we randomly assigned high-risk patients with atrial fibrillation in a 1:1 ratio to receive asundexian at a dose of 50 mg once daily or standard-dose apixaban.

Gender and contemporary risk of adverse events in atrial fibrillation.

Eur Heart J

September 2024

Institute of Cardiovascular Sciences, University of Birmingham, Medical School, Vincent Drive, Birmingham B15 2TT, UK.

Article Synopsis
  • This study investigates how gender affects decision-making for oral anticoagulation in patients with atrial fibrillation (AF), using a large dataset of over 16 million patients from UK primary care between 2005-2020.
  • It found that in patients aged 40-75 without prior strokes, women had a lower adjusted rate of primary outcomes (death, ischemic stroke, or thromboembolism) compared to men, primarily due to lower mortality rates in women.
  • The study concludes that omitting gender from clinical risk scores could streamline the process of determining which AF patients should receive oral anticoagulation.

Article Synopsis
  • The study investigates the relationship between serum calcium levels and ventricular repolarization time, specifically the QT and JT intervals, which are important for heart health.
  • Researchers conducted large-scale genome-wide analyses to explore potential interactions between calcium levels and genetic variants associated with QT and JT intervals, using over 122,000 participants.
  • The results showed limited evidence for the hypothesized calcium interaction effects, suggesting that other factors, such as rare genetic variations or environmental influences, likely play a larger role in the unexplained heritability of QT and JT intervals.

Lipids contribute to hematopoiesis and membrane properties and dynamics; however, little is known about the role of lipids in megakaryopoiesis. Here we show that megakaryocyte progenitors, megakaryocytes and platelets present a unique lipidome progressively enriched in polyunsaturated fatty acid (PUFA)-containing phospholipids. In vitro, inhibition of both exogenous fatty acid functionalization and uptake as well as de novo lipogenesis impaired megakaryocyte differentiation and proplatelet production.
