In silico toxicology (IST) approaches to rapidly assess chemical hazard, and usage of such methods is increasing in all applications but especially for regulatory submissions, such as for assessing chemicals under REACH as well as the ICH M7 guideline for drug impurities. There are a number of obstacles to performing an IST assessment, including uncertainty in how such an assessment and associated expert review should be performed or what is fit for purpose, as well as a lack of confidence that the results will be accepted by colleagues, collaborators and regulatory authorities. To address this, a project to develop a series of IST protocols for different hazard endpoints has been initiated and this paper describes the genetic toxicity in silico (GIST) protocol.

Fragrance allergens in household detergents.

Octanol-Water Partition Coefficient Measurement by a Simple H NMR Method.

We describe a simple miniature shake-flask method to measure the octanol-water partition coefficient of an organic compound. Partition between water and octanol is performed in an NMR tube; the aqueous phase is analyzed by H NMR spectroscopy using a benchtop low-field NMR instrument. Neither pre-equilibration of solvents nor isolation of the two phases is required.

For the first time the occurrence of 26 nitrogen heterocyclic polyaromatic hydrocarbons (N-HETs) and the antiepileptic drug carbamazepine (CBZ) as a marker for anthropogenic influence was determined in an entire river system and its influents. Besides the investigation of diurnal and annual variations in concentrations, dilution and load calculations, the aims were to facilitate transferability to other rivers by identifying sources of the different substances. To create a sufficient database over 180 aqueous samples from the River Fuhse (Lower Saxony, Germany), its catchment area, effluents of municipal and industrial wastewater treatment plants (WWTPs) and rainwater were analyzed.

Engineered nanoparticle (ENP) fate models developed to date - aimed at predicting ENP concentration in the aqueous environment - have limited applicability because they employ constant environmental conditions along the modeled system or a highly specific environmental representation; both approaches do not show the effects of spatial and/or temporal variability. To address this conceptual gap, we developed a novel modeling strategy that: 1) incorporates spatial variability in environmental conditions in an existing ENP fate model; and 2) analyzes the effect of a wide range of randomly sampled environmental conditions (representing variations in water chemistry). This approach was employed to investigate the transport of nano-TiO2 in the Lower Rhône River (France) under numerous sets of environmental conditions.

The use of redox potential measurements for corrosion and scaling monitoring, including microbially mediated processes, is demonstrated. As a case study, monitoring data from 10years of operation of an aquifer thermal energy storage (ATES) site located in Berlin, Germany, were examined. (Fe(2+))-activities as well as [Fe(3+)]-build up rates were calculated from redox potential, pH, conductivity, temperature and dissolved oxygen measurements.
