Residual dipolar couplings (RDCs) and other residual anisotropic NMR parameters provide valuable structural information of high quality and quantity, bringing detailed structural models of flexible molecules in solution in reach. The corresponding data interpretation so far is directly or indirectly based on the concept of a molecular alignment tensor, which, however, is ill-defined for flexible molecules. The concept is typically applied to a single or a small set of lowest energy structures, ignoring the effect of vibrational averaging.

The potential of residual dipolar couplings (RDCs) in conformational studies of small molecules is now widely recognized, but current theoretical approaches for their interpretation have several limitations and there is still the need for a general method to probe the torsional angle distributions applicable to any rotationally flexible molecule. Molecular dynamics simulations with RDC-based orientational tensorial constraints (MDOC), implemented in the software COSMOS, are presented here as a conceptually new strategy. For the cases of the fluorinated anti-inflammatory drug diflunisal and the disaccharide cellobiose, we demonstrate that MDOC simulations with one-bond RDCs as tensorial constraints unveil torsion distributions and allow the determination of relative configuration in the presence of rotational flexibility.

Intramembrane cleavage of the β-amyloid precursor protein C99 substrate by γ-secretase is implicated in Alzheimer's disease pathogenesis. Biophysical data have suggested that the N-terminal part of the C99 transmembrane domain (TMD) is separated from the C-terminal cleavage domain by a di-glycine hinge. Because the flexibility of this hinge might be critical for γ-secretase cleavage, we mutated one of the glycine residues, G38, to a helix-stabilizing leucine and to a helix-distorting proline.

Nuclear magnetic resonance (NMR) spectroscopy is one of the most promising methods for use in metabolomics studies as it is able to perform non targeted measurement of metabolites in a quantitative and non-destructive way. Sample preparation of liquid samples like urine or blood serum is comparatively easy in NMR metabolomics, because mainly buffer and chemical shift reference substance are added. For solid samples like feces suitable extraction protocols need to be defined as initial step, where the exact protocol depends on sample type and features.

Numerically optimised microwave pulses are used to increase excitation efficiency and modulation depth in electron spin resonance experiments performed on a spectrometer equipped with an arbitrary waveform generator. The optimisation procedure is sample-specific and reminiscent of the magnet shimming process used in the early days of nuclear magnetic resonance - an objective function (for example, echo integral in a spin echo experiment) is defined and optimised numerically as a function of the pulse waveform vector using noise-resilient gradient-free methods. We found that the resulting shaped microwave pulses achieve higher excitation bandwidth and better echo modulation depth than the pulse shapes used as the initial guess.

Lignocellulose can be converted sustainably to fuels, power and value-added chemicals like fatty acid esters. This study presents a concept for the first eco-friendly enzymatic synthesis of economically important fatty acid sugar esters based on lignocellulosic biomass. To achieve this, beech wood cellulose fiber hydrolysate was applied in three manners: as sugar component, as part of the deep eutectic solvent (DES) reaction system and as carbon source for the microbial production of the fatty acid component.

Background: Deficient glucocorticoid biosynthesis leading to adrenal insufficiency is life-threatening and is associated with significant co-morbidities. The affected pathways underlying the pathophysiology of co-morbidities due to glucocorticoid deficiency remain poorly understood and require further investigation.
Methods: To explore the pathophysiological processes related to glucocorticoid deficiency, we have performed global transcriptional, post-transcriptional and metabolic profiling of a cortisol-deficient zebrafish mutant with a disrupted ferredoxin () system.

The borane-catalyzed synthesis of quinoline derivatives bearing tetrasubstituted stereocenters from vinyl anilines has been developed. Mechanistic studies and quantum-mechanical investigations support the hydride abstraction/electrocyclization/hydride addition mechanism. The products were obtained in up to 99 % yield with a diastereoselectivity of >99 % in favour for the 3a-5-cis isomer.

The controlled incorporation of phenanthroline moieties into polymers is introduced, demonstrating their application as metal-ion complexing ligands for the construction of advanced macromolecular structures. Specifically, two phenanthroline-containing monomers based on acrylate and styrene functionalities, were synthesized. Each monomer was readily copolymerized with either N,N-dimethylacrylamide or styrene via nitroxide-mediated polymerization, resulting in narrowly distributed polar or non-polar copolymers.

Similar to J-resolved spectroscopy, also, heteronuclear multiple bond correlation (HMBC), heteronuclear single bond correlation (HSBC), and heteronuclear multiple quantum coherence (HMQC) types of correlation experiments result in homonuclear tilted multiplet patterns. On the example of the high-resolution heteronuclear single bond correlation (HR-HSBC) pulse sequence, it is shown how the tilt angle can be varied within a wide range of positive and negative values. Projection along the tilt angles in all cases results in homonuclear decoupling.

Novel CLIP-COSY based homo- and heteronuclear correlation experiments are reported for the rapid, semi-automated NMR assignment of small to medium-sized molecules. The homonuclear CLIP-COSY and corresponding relayed experiments employ the perfect-echo based mixing sequence for in-phase coherence transfer between directly and/or indirectly coupled proton spins. The combined analysis of the resulting CLIP-COSY and relayed spectra made it possible to easily track down, layer by layer, the proton-proton connectivity network.

The fungal pathogen Fusarium pseudograminearum causes important diseases of wheat and barley. During a survey of secondary metabolites produced by this fungus, a novel class of cytokinins, herein termed Fusarium cytokinins, was discovered. Cytokinins are known for their growth-promoting and anti-senescence activities, and the production of a cytokinin mimic by what was once considered as a necrotrophic pathogen that promotes cell death and senescence challenges the simple view that this pathogen invades its hosts by employing a barrage of lytic enzymes and toxins.

Previously we introduced two novel NMR experiments for small molecules, the so-called ASAP-HSQC and ALSOFAST-HSQC (Schulze-Sünninghausen et al., 2014), which allow the detection of heteronuclear one-bond correlations in less than 30s at natural abundance. We propose an improved symmetrized pulse scheme of the basic experiment to minimize artifact intensities and the combination with non-uniform sampling to enable the acquisition of conventional HSQC spectra in as short as a couple of seconds and extremely C-resolved spectra in less than ten minutes.
