The application of upconversion nanoparticles (UCNPs) for cell and tissue analysis requires a comprehensive understanding of their interactions with biological entities to prevent toxicity or harmful effects. Whereas most studies focus on cancer cells, this work addresses non-cancerous cells with their regular in vitro physiology. Since it is generally accepted that surface chemistry largely determines biocompatibility in general and uptake of nanomaterials in particular, two bilayer surface coatings with different surface shielding properties have been studied: (i) a phospholipid bilayer membrane (PLM) and (ii) an amphiphilic polymer (AP).

Plant triterpenoids represent a diverse group of secondary metabolites and are thought to be valuable for therapeutic applications. For drug development, lead optimization, better knowledge of biological pathways, and high-throughput detection of secondary metabolites in plant extracts are crucial. This paper describes a qualitative method for the rapid and accurate identification of various triterpenoids in plant extracts using the LC-HR-ESI-MS/MS tool in combination with the data-dependent acquisition (DD) approach.

Tire and road wear particles (TRWP) are a major contributor to non-exhaust traffic emissions, but their contribution to and dynamics in urban aerosol is not well known. Urban particulate matter (PM) in the size fraction below 10 µm (PM) from two German cities was collected over 2 weeks and analysed for 39 tire-related chemicals, including amines, guanidines, ureas, benzothiazoles, p-phenylenediamines, quinolines and several transformation products (TPs). Of these, 37 compounds were determined in PM at median concentrations of 212 pg/m for 1,3-diphenylguanidine (DPG) and 132 pg/m for benzothiazole-2-sulfonic acid (BTSA); 10 of the compounds have not been reported in urban aerosol before.

Internal concentrations (ICs) are crucial for linking exposure to effects in the development of New Approach Methodologies. ICs of chemicals in aquatic organisms are primarily driven by hydrophobicity and modulated by biotransformation and efflux. Comparing the predicted baseline to observed toxicity enables the estimation of effect specificity, but biological processes can lead to overestimating ICs and bias the specificity assessment.

Persistent and mobile (PM) chemicals are considered detrimental for drinking water resources as they may pass through all barriers protecting these resources against pollution. However, knowledge on the occurrence of PM chemicals in the water cycle, that make their way into drinking water resources, is still limited. The effluents of six municipal wastewater treatment plants (WWTPs, n = 38), surface water of two rivers (n = 32) and bank filtrate of one site (n = 15) were analyzed for 127 suspected PM chemicals.

Using ultrafast spectroscopy and site-specific mutagenesis, we demonstrate the central role of a conserved tyrosine within the chromophore binding pocket in the forward (P → P) photoconversion of phytochromes. Taking GAF1 of the knotless phytochrome All2699g1 from Nostoc as representative member of phytochromes, it was found that the mutations have no influence on the early (<30 ps) dynamics associated with conformational changes of the chromophore in the excited state. Conversely, they drastically impact the extended protein-controlled excited state decay (>100 ps).

A putative xanthorhodopsin-encoding gene, XR34, was found in the genome of the moderately halophilic gammaproteobacterium Salinivibrio socompensis S34, isolated from modern stromatolites found on the shore of Laguna Socompa (3570 m), Argentina Puna. XR-encoding genes were clustered together with genes encoding X-carotene, retinal (vitamin-A aldehyde), and carotenoid biosynthesis enzymes while the carotene ketolase gene critical for the salinixanthin antenna compound was absent. To identify its functional behavior, we herein overexpressed and characterized this intriguing microbial rhodopsin.
