Introduction: Cervical cancer is a public health issue in Africa with devastating socioeconomic consequences due to the lack of organized screening programs. The success of screening programs depends on the appropriate investigation and management of women who test positive for screening. Colposcopic assessment following positive screening results is a noteworthy issue in Africa.

December 2024

Division of Pharmacogenomics and Personalized Medicine, Department of Pathology, Faculty of Medicine, Ramathibodi Hospital, Mahidol University, Bangkok, 10400, Thailand.

DPYD polymorphisms have been widely found to be related to 5-FU-induced toxicities. The aim of this study was to establish significant associations between five single-nucleotide polymorphisms of DPYD and 5-FU hematological toxicities in Thai colorectal cancer patients. The toxicities were analyzed at the first and second cycles of 5-FU administration in 75 patients.

Transplant in myeloma: who, when, and why?

Hematology Am Soc Hematol Educ Program

December 2024

Hematology Department, Toulouse University Hospital, Institut Universitaire du Cancer de Toulouse-Oncopole CHU Toulouse, Toulouse, France.

High-dose melphalan supported by autologous transplantation has been the standard of care for eligible patients with newly diagnosed multiple myeloma for nearly 30 years. Several randomized clinical trials have reaffirmed the strong position of transplant in the era of triplets combining proteasome inhibitors, immunomodulatory drugs, and dexamethasone. Although quadruplets are becoming the standard in transplantation programs, no data are currently available on the need for a transplant with new regimens incorporating anti-CD38 monoclonal antibodies.

Introduction: Sickle cell disease (SCD) is one of the most common genetic diseases in the world, annually affecting approximately 310 000 births and causing >100 000 deaths. Vaso-occlusive crisis (VOC) is the most frequent complication of SCD, leading to bone pain, thoracic pain (acute chest syndrome) and/or abdominal spasms. It is the main cause of mortality in patients with SCD, reducing life expectancy.

Unlabelled: This phase 2 study investigated pevonedistat + azacitidine + venetoclax ( = 83) versus azacitidine + venetoclax ( = 81) in patients with newly diagnosed acute myeloid leukemia (AML) ineligible for intensive chemotherapy. The study was stopped early following negative results from PANTHER, which evaluated pevonedistat in higher-risk myelodysplastic syndromes/chronic myelomonocytic leukemia or low-blast AML. Outcomes were analyzed up to the datacut.

Article Synopsis
  • The study aimed to investigate the relationship between the immune microenvironment and survival outcomes in patients with vulvovaginal melanoma.
  • It involved a retrospective analysis of 42 patients, examining tumor-infiltrating lymphocytes and genetic mutations, with significant findings on immune cell presence related to better disease-free survival.
  • The researchers concluded that specific types of tumor-infiltrating lymphocytes could indicate disease progression and response to treatments, suggesting the need for further multicenter studies to confirm these results.

The aim of our study was to analyze the potential survival benefit associated with HSCT according to clinico-biological scores which incorporate molecular data (MIPSS70 and MIPSS70+V2) to facilitate decision-making in this context. One transplant (n=241) and one non-transplant cohorts (n=239) were used to test the hypothesis that PMF patients with higher risk molecular score benefit from HSCT. A weighted propensity score was applied to balance confounding factors with the transplanted cohort as reference.

According to current recommendations, older AML patients in first complete remission (CR) after induction chemotherapy should receive consolidation with intermediate-dose cytarabine (IDAC). However, no study has demonstrated the superiority of IDAC over other regimen. In this retrospective study, we compared the efficacy of mini-consolidations (idarubicin 8 mg/m day 1, cytarabine 50 mg/m/12 h, day 1-5) and IDAC.

November 2024

Salmonella and Listeria Unit (SEL), ANSES, Laboratory for Food Safety, Maisons-Alfort, France.

Article Synopsis
  • Typhimurium and its monophasic variant (4,[5],12:i:-) are common serovars, but their spread in French pig herds and slaughterhouses has not been previously explored at a regional level.
  • The study analyzed 188 strains from French pigs collected between 2014 and 2019, revealing limited genomic diversity, indicating that only a couple of clones are prevalent among pig populations.
  • By comparing these strains with international isolates, the research highlights specific genomic characteristics of the ST 34 monophasic variants in France, along with other findings related to virulence and antimicrobial resistance.

Midostaurin shapes macroclonal and microclonal evolution of FLT3-mutated acute myeloid leukemia.

Blood Adv

October 2024

CHU Bordeaux, Service d'Hématologie Clinique et Thérapie cellulaire, F-33000, Bordeaux, France, PESSAC, France.

Article Synopsis
  • Despite initial treatment with midostaurin (MIDO) and chemotherapy in FLT3-mutated acute myeloid leukemia (AML), many patients face relapses, with complete remission rates around 60-70% and over 40% relapsing.
  • A study of 150 patients with refractory/relapsed (R/R) AML revealed that those treated with MIDO showed lower persistence of FLT3-ITD mutations compared to those who did not receive MIDO (68% vs. 87.5%).
  • The study found that detecting multiple FLT3-ITD clones at diagnosis related to a higher persistence rate of these mutations at relapse, indicating the need for sensitive techniques in FLT3-
