JAK inhibition for CD3 CD4 lymphocytic-variant hypereosinophilic syndrome.

Alternatives are urgently needed in patients with CD3 CD4 lymphocytic-variant hypereosinophilic syndrome (L-HES) requiring high-level steroids or who are unresponsive and/or intolerant to conventional alternative therapies. We report five L-HES patients (44-66 years) with cutaneous involvement (n = 5) and persistent eosinophilia (n = 3) despite conventional therapies, who successfully received JAK inhibitors (tofacitinib n = 1, ruxolitinib n = 4). JAKi led to complete clinical remission in the first 3 months in all (with prednisone withdrawal in four).

The escalating burden of antibiotic drug resistance necessitates research into novel classes of antibiotics and their mechanism of action. Pyrrolomycins are a family of potent natural product antibiotics with nanomolar activity against Gram-positive bacteria, yet with an elusive mechanism of action. In this work, we dissect the apparent Gram-positive specific activity of pyrrolomycins and show that Gram-negative bacteria are equally sensitive to pyrrolomycins when drug efflux transporters are removed and that albumin in medium plays a large role in pyrrolomycin activity.

Intrinsically disordered protein YAP (yes-associated protein) interacts with TEADs transcriptional factors family (transcriptional enhancer associated domain) creating three interfaces. Interface 3, between the Ω-loop of YAP and a shallow pocket of TEAD was identified as the most important TEAD zone for YAP-TEAD interaction. Using the first X-ray structure of the hYAP-hTEAD1 complex (PDB 3KYS) published in 2010, a protein-protein interaction inhibitors-enriched library (175,000 chemical compounds) was screened against this hydrophobic pocket of TEAD.

  • IVIG (intravenous immunoglobulins) may be a beneficial treatment for patients with systemic sclerosis (SSc) due to their immunomodulatory and antifibrotic effects.
  • A study involving 46 SSc patients showed significant improvements in muscle pain, muscle weakness, and joint pain after IVIG treatment, along with reductions in inflammatory markers and corticosteroid usage.
  • The treatment was generally safe, with few severe side effects reported, suggesting that IVIG could effectively manage symptoms and decrease reliance on corticosteroids in SSc patients.
