Takeda G-protein-coupled receptor 5 (TGR5) is emerging as an important and promising target for the development of anti-diabetic drugs. To understand the structural characteristics of TGR5 agonists, the common feature pharmacophore models were generated and molecular docking was performed. Objective: To evaluate the effects of topical oxygen therapy and its impacts on granulation tissue in patients with chronic traumatic wounds.
Method: A total of 112 patients with chronic traumatic wounds were randomly divided into the control group (n=56, receiving negative-pressure wound therapy) and the intervention group (n=56, receiving negative-pressure wound therapy plus topical oxygen therapy) using a random number table and they were treated continuously for 2 weeks. Then, the scores from the Pressure Ulcer Scale for Healing (PUSH), the coverage rate of granulation tissue, the severity of pain and Transcutaneous Oxygen Partial Pressure (TcPO) before and after treatment were compared between the two groups.

Farnesoid X receptor (FXR) agonists would be considered as an important therapeutic strategy for several chronic liver and metabolic diseases. Here we have employed an integrated virtual screening by combining ligand-based pharmacophore mapping and molecular docking to identify novel nonsteroidal FXR agonists. Eighteen compounds were selected for FXR agonistic activity assay, and results showed five compounds exhibiting promising FXR agonistic activity.

Five Mn(i) photo-activated carbon monoxide-releasing molecules (photo-CORMs) with benzimidazole coligands, namely [MnBr(CO)L1] (1, L1 = 2-(2-pyridyl)benzimidazole), [Mn(CO)L1(PPh)](ClO) (2), [MnBr(CO)L2] (3, L2 = 2,2'-bisbenzimidazole), [MnBr(CO)L3]·CHOH (4, L3 = 2,6-bis(benzimidazole-2'-yl)pyridine) and -[MnBr(CO)L4] (5, L4 = 2,4-bis(benzimidazole-2'-yl) pyridine) were synthesized by reactions of MnBr(CO) with complexes L1-L4, respectively, and characterized single crystal X-ray diffraction, elemental analysis, H-NMR, C-NMR, IR, UV-vis and fluorescence spectroscopy. The CO-release properties of 1-5 were investigated using the myoglobin assay and CO detection, and the results show that all of the complexes could release CO rapidly upon exposure to 365 nm UV light. Comparing their half-lives of CO release, we found that increasing the degree of unsaturation and conjugation of the ligand frame could be advantageous for prolonging the time of CO-release, and that the luminescence intensity of 1-5 could gradually be enhanced.
