Immunology

November 2023

Jiangsu Key Laboratory for Molecular and Medical Biotechnology, College of Life Sciences, Nanjing Normal University, Nanjing, China.

Article Synopsis
  • CAR-T-cell immunotherapy shows great promise for blood cancers but struggles with solid tumors due to inefficient T-cell infiltration.
  • Flap structure-specific endonuclease 1 (FEN1) plays a crucial role in DNA repair and is often overexpressed in cancer; inhibiting it may enhance cancer treatment.
  • Combining FEN1 inhibitors like SC13 with CAR-T-cell therapy increases double-stranded DNA in tumor cells, which boosts chemokine secretion and helps CAR-T cells effectively infiltrate solid tumors, improving anti-tumor immunity.

October 2023

Jiangsu Key Laboratory for Molecular and Medical Biotechnology, College of Life Sciences, Nanjing Normal University, 1 WenYuan Road, Nanjing, 210023, China.

Article Synopsis
  • CAR-T cell immunotherapy has seen success in treating blood cancers, but is challenging for solid tumors due to difficulty in T cell infiltration and long-term immune response.
  • Dendritic cells (DCs) can improve this by presenting tumor antigens and enhancing T cell infiltration, making the combination of DC vaccines and CAR-T therapy a promising treatment strategy for solid tumors.
  • Research showed that DC vaccines boosted MSLN CAR-T cell proliferation, infiltration, and persistence in mouse models, suggesting a potential pathway for more effective CAR-T therapies in clinical settings.

Clinical Medical Research Center of the Affiliated Hospital/Inner Mongolia Key Laboratory of Medical Cellular Biology, Inner Mongolia Medical University, Hohhot, 010050, Inner Mongolia, P.R. China.

MicroRNAs (miRNAs or miRs) is a non-coding small RNA of a type of 18~24 nucleotide-regulated gene that has been discovered in recent years. It mainly degrades the target gene mRNA or inhibits its translation process through the complete or incomplete bindings with 3'UTR of target genes, followed by the regulation of individual development, apoptosis, proliferation, differentiation and other life activities through the post-transcriptional regulation. Among many miRNAs, the microRNA family, miR-30, plays diverse roles in these key process of neoplastic transformation, metastasis, and clinical outcomes in different cancer progression.
