For confirmatory trials of regulatory decision making, it is important that adaptive designs under consideration provide inference with the correct nominal level, as well as unbiased estimates, and confidence intervals for the treatment comparisons in the actual trials. However, naive point estimate and its confidence interval are often biased in adaptive sequential designs. We develop a new procedure for estimation following a test from a sample size reestimation design.

The electrical properties of the anterior stomach of the larval mosquito (Aedes aegypti) were determined. At late times after cannulation, the intraluminal space constant was 936 microm, which is almost as long as the isolated tissue itself. At this time, the resistance of the apical cell membranes dominates the transcellular resistance; it is approximately 14 times the resistance of the basal cell membrane.

A new fluorescent indicator for metal ions, called Constant Calcein Blue, has been synthesized by condensing 7-hydroxy-2,3-dimethylchromone, iminodiacetic acid and formaldehyde. The indicator exhibits extraordinary resistance to alkaline hydrolysis and is recommended for spectrofluorometric determinations of metal ions, such as calcium, that must be performed at pH 10.5 and higher.

Studies of 7-hydroxybenzo-4-pyrones.

Spectral data and rates of hydrolysis in 0.7M potassium hydroxide are reported for a series of nine 7-hydroxybenzo-4-pyrones differing in substitution at positions 2 and 3. The results reveal that 2,3-dimethyl substitution produces a molecule, 7-hydroxy-2,3-dimethylchromone, sufficiently fluorescent and resistant to alkaline hydrolysis to warrant its use as the fluorescent moiety in a metallofluorescent indicator of the Calcein Blue type.
