Bicalutamide (Bic) is an androgen deprivation therapy (ADT) for treating prostate cancer, while ADT is potentially associated with acute kidney injury. Previously, we recognized Bic induced renal mitochondria dysfunction in vitro and in vivo via the ROS -HIF1α pathway. Whether OXPHOS complex, as well as mitochondrial dynamics, can be influenced by Bic via modulation of peroxisome proliferator-activated receptor coactivator 1α (PGC1α), NADPH oxidase 4 (Nox4), mitofusins 1/2 (MFN 1/2), optic atrophy 1 (OPA1), and sirtuins (SIRTs) has not been documented.

The protective effects of water extracts of djulis () (WECF) and their bioactive compounds on particulate matter (PM)-induced oxidative injury in A549 cells via the nuclear factor-erythroid 2-related factor 2 (Nrf2) signaling were investigated. WECF at 50-300 µg/mL protected A549 cells from PM-induced cytotoxicity. The cytoprotection of WECF was associated with decreases in reactive oxygen species (ROS) generation, thiobarbituric acid reactive substances (TBARS) formation, and increases in superoxide dismutase (SOD) activity and glutathione (GSH) contents.

Article Synopsis
  • Cinnamon is a traditional Chinese medicine that is used to treat nocturia and may help alleviate overactive bladder (OAB) symptoms without significant side effects, but its mechanism of action is not fully understood.
  • In a clinical study, cinnamon powder patches were effective in reducing OAB symptoms in patients, and in a murine model of OAB induced by cyclophosphamide (CYP), treatment with cinnamon powder (CNP) and its active component cinnamaldehyde (CNA) significantly improved urination patterns and bladder pathology.
  • The study found that CNP and CNA treatments suppressed inflammation and fibrosis pathways associated with bladder dysfunction compared to the clinical drug solifenacin, suggesting that cinnamon may offer a beneficial alternative for OAB

Pulmonary arterial hypertension (PAH) is characterized by muscularized pulmonary blood vessels, leading to right heart hypertrophy and cardiac failure. However, state-of-the-art therapeutics fail to target the ongoing remodeling process. Here, this study shows that matrix metalloproteinases (MMP)-1 and MMP-10 levels are increased in the medial layer of vessel wall, serum, and M1-polarized macrophages from patients with PAH and the lungs of monocrotaline- and hypoxia-induced PAH rodent models.

Vascular impairment is a crucial factor associated with chronic muscle pain, but relevant research from the microcirculatory aspect is lacking. Here, we investigated the differences in neck muscle microcirculation detected through laser-doppler flowmetry (LDF) and cervical biomechanics by a videofluoroscopic image in asymptomatic participants and patients with postural neck and shoulder pain. To understand the mechanism behind the effect of myofascial treatment, transverse friction massage (TFM) was applied and the immediate effects of muscular intervention on microcirculation were monitored.

Article Synopsis
  • Doxorubicin (Dox) is a common cancer treatment but can lead to muscle wasting and other adverse effects, impacting patients' quality of life.
  • The study investigated the effects of low-level laser (LLL) therapy on muscle protection in rat and C2C12 myoblast models subjected to Dox, finding that LLL significantly reduces Dox-induced muscle loss and mitochondrial damage.
  • The protective effects of LLL were linked to the activation of the AMPK/SIRT1/PGC-1α signaling pathway, suggesting that LLL therapy could be a promising adjunct treatment for mitigating Dox-related muscle toxicity in cancer patients.

Article Synopsis
  • A study investigated the effectiveness of modifying a triangular configuration for fixing transverse patellar fractures by replacing some cannulated screws with headless compression screws (HCSs).
  • Finite element simulations were performed to compare the mechanical stability of various configurations, including combinations of HCSs and cannulated screws, under different knee positions and loading conditions.
  • The results showed that using three HCSs in a backward triangular configuration provided the best stability, significantly reducing fragment displacement and fracture gap size compared to traditional methods with cannulated screws and anterior wiring.

Renal tubulointerstitial lesions (TILs), a common pathologic hallmark of chronic kidney disease that evolves to end-stage renal disease, is characterized by progressive inflammation and pronounced fibrosis of the kidney. However, current therapeutic approaches to treat these lesions remain largely ineffectual. Previously, we demonstrated that elevated IL-36α levels in human renal tissue and urine are implicated in impaired renal function, and IL-36 signaling enhances activation of NLRP3 inflammasome in a mouse model of TILs.
