Obesity is a risk factor for postmenopausal breast cancer (BC), and evidence suggests a role for adiponectin in the relationship between obesity and BC. We investigated whether adiponectin or other biomarkers mediate the effect of body mass index (BMI) on postmenopausal BC risk in a cohort study nested in the IBIS-II Prevention Trial. We measured adiponectin, leptin, IGF-I, IGFBP-1, high-sensitivity C-reactive protein, glycemia, insulin, HOMA-IR index, and SHBG in baseline and 12-month serum samples from 123 cases and 302 matched controls in the placebo arm of the IBIS-II Prevention trial.

Arterial hypertension represents the most important cardiovascular risk factor with a direct responsibility for a large share of cardiovascular mortality and morbidity in the world. Despite the wide availability of antihypertensive therapies with documented effectiveness, blood pressure control still remains largely unsatisfactory in large segments of the population. Guidelines for the management of arterial hypertension suggest the preferential use of five classes of drugs-angiotensin-converting enzyme inhibitors, angiotensin II type I receptor inhibitors, calcium channel blockers, thiazide/thiazide-like diuretics, and beta-blockers-recommending the use of combination therapy, preferably in pre-established combinations, for the majority of hypertensive patients.

Article Synopsis
  • The study investigated how smoking status affects the coronary volume-to-myocardial mass ratio (V/M) in individuals with coronary artery disease (CAD) who underwent CT analysis.
  • It included a sample of 2,874 participants, revealing that former smokers had higher coronary volume than never-smokers, while current smokers had greater myocardial mass but both groups exhibited lower V/M ratios compared to never-smokers.
  • The findings suggest that both current and former smoking status are significant predictors of lower V/M, alongside other factors like diabetes and severity of coronary stenosis.

Article Synopsis
  • - Microglia and macrophages produce extracellular vesicles (EVs) that can affect the development of myelin lesions, with microglial EVs supporting oligodendrocyte precursor cell (OPC) differentiation, while the effect of macrophage EVs is less clear.
  • - Research showed that EVs from macrophages, especially those in an inflammatory state, do not aid OPC differentiation and actually inhibit it, but their lipid content can still promote OPC maturation similar to microglial EVs.
  • - Key promyelinating endocannabinoids (like anandamide) were found in EVs from both cell types, indicating a critical role for these endocannabinoids in OPC maturation and suggesting potential targets for enhancing myelin repair

Article Synopsis
  • - The study investigates whether pathologic complete response (pCR) and major pathologic response (MPR) can reliably predict event-free survival (EFS) and overall survival (OS) in neoadjuvant trials for non-small cell lung cancer (NSCLC).
  • - A total of seven clinical trials involving 2385 patients were analyzed, demonstrating a strong correlation between pCR/MPR and 2-year EFS, but weaker associations with OS.
  • - The conclusion suggests that while pCR and MPR are good indicators of EFS, their predictive value for OS is less clear; further research is needed for more accurate data and understanding of patient outcomes.

The translocation of the testis-determining factor, the SRY gene, from the Y to the X chromosome is a rare event that causes abnormalities in gonadal development. In all cases of males and females carrying this translocation, disorder of sex development is reported. In our study, we described a peculiar pedigree with the first evidence of four healthy females from three generations who are carriers of the newly identified t(X;Y)(q28;p11.

Article Synopsis
  • Immune thrombocytopenia (ITP) leads to low platelet counts due to increased destruction and is classified based on the time post-diagnosis: newly diagnosed, persistent, and chronic.
  • The TAPER trial studied eltrombopag as a treatment option for adults with ITP who didn't respond to corticosteroids, finding that about 30.5% of participants achieved a sustained response off-treatment after a year.
  • Eltrombopag improved platelet counts and quality of life within a few months, with a median duration of sustained response extending to about 22 months in later follow-ups, while maintaining a safety profile consistent with prior studies.

The tradeoff between speed and accuracy is a well-known constraint for human movement, but previous work has shown that this tradeoff can be modified by practice, and the quantitative relationship between speed and accuracy may be an indicator of skill in some tasks. We have previously shown that children with dystonia are able to adapt their movement strategy in a ballistic throwing game to compensate for increased variability of movement. Here, we test whether children with dystonia can adapt and improve skills learned on a trajectory task.
