Plant receptor kinases (RKs) are critical for transmembrane signalling involved in various biological processes including plant immunity. MALE DISCOVERER1-INTERACTING RECEPTOR-LIKE KINASE 2 (MIK2) is a unique RK that recognizes a family of immunomodulatory peptides called SERINE-RICH ENDOGENOUS PEPTIDEs (SCOOPs) and activates pattern-triggered immunity responses. However, the precise mechanisms underlying SCOOP recognition and activation of MIK2 remain poorly understood.

State Key Laboratory of Advanced Chemical Power Sources, Frontiers Science Center for New Organic Matter, Key Laboratory of Advanced Energy Materials Chemistry (Ministry of Education), College of Chemistry, Nankai University, Tianjin, 300071, P. R. China.

Pure-halide reduced-dimensional perovskites, featuring large exciton binding energy and tunable bandgap, show great potential for high-efficiency deep-blue perovskite light-emitting diodes (PeLEDs). However, their efficiency, particularly in the low n-value phase domain ("n" represents the number of octahedral sheets), lags behind analogous perovskite emitters. Herein, it is demonstrated that the vibration of edge-dangling octahedra in the low n-value phase activates notorious exciton-phonon (EP) coupling, thereby deteriorating efficiency.

Isoindolones constitute a dominant structural class in synthetic and medicinal chemistry. In this research, an electrochemical reaction involving 2-alkynylbenzamides, inorganic sulfites, and alcohols was first established to provide sulfonyl ester-substituted 3-hydroxyisoindolinone derivatives in moderate to good yields with excellent functional group tolerance. When bulky aryl-substituted 2-alkynylbenzamides are utilized as substrates, sulfonyl ester-substituted 3-alkylideneisoindolinones can be selectively generated with good chemoselectivity.

Amonafide (ANF), a topoisomerase II inhibitor and DNA intercalator, has exhibited promise in phase II trials but faces significant limitations due to adverse side effects. Here, we have developed a novel enzyme-triggered fluorogenic prodrug, AcKLP, that incorporates dual-locked enzyme activation, ensuring that the prodrug remains inactive until it confronts the unique enzymatic environment of glioblastoma cells. This approach minimizes premature activation and reduces toxicity to normal cells, with an IC > 100 μM for human umbilical vein endothelial cells (HUVEC) and ∼2.

J Org Chem

November 2024

School of Petrochemical Engineering, Institute of Functional Heterocycles, Changzhou University, Changzhou 213164, China.

A Bro̷nsted acid-catalyzed [5 + 1] annulation of 2-(1-indol-2-yl)phenols, a class of indole-based 1,5-CO-synthons, with aldehydes has been established. By this approach, various indole-fused dihydrochromanes were synthesized in moderate to good yields (up to 99%) with excellent regioselectivities. Control experiments not only showed the H-bonding interaction between the NH group of the substrates─and the acid catalyst is vital to realize this transformation─but also indicated that an indolylmethanol-type intermediate was possibly formed via the first step of nucleophilic addition of indole C3-position to aldehydes during the reaction process.

A visible-light induced intermolecular hydroamination of alkenes with azoles is reported, delivering pharmaceutically valuable N-benzyl azoles in high yields with excellent Markovnikov selectivity. Mechanistic studies suggest that the process is initiated by the energy transfer of the excited photocatalyst with alkenes, followed by the single electron reduction, protonation, and subsequent single electron oxidation to afford the key alkyl carbocation intermediate. This protocol exhibits advantages of broad functional group tolerance, excellent atom economy, high efficiency, and mild reaction conditions.
