Article Synopsis
  • The study used Optical Coherence Tomography (OCT) to assess the thickness of retinal layers in people with epilepsy, categorizing them into drug-resistant and non-drug-resistant groups compared to healthy controls.
  • Results showed that both epilepsy groups had thinner retinal layers than controls, with the drug-resistant group having significantly reduced ganglion cell-inner plexiform layer thickness.
  • These findings indicate that drug resistance in epilepsy is linked to changes in retinal structure, suggesting potential biomarkers for epilepsy management.

Real-world smartphone data can trace the behavioural impact of epilepsy: A case study.

Eur J Neurol

December 2024

Stichting Epilepsie Instellingen Nederland (SEIN), Heemstede, The Netherlands.

Background: Neurobehavioural comorbidities have a detrimental effect on the quality of life of people with epilepsy, yet tracking their impact is challenging as behaviour may vary with seizures and anti-seizure medication (ASM) side effects. Smartphones have the potential to monitor day-to-day neurobehavioural patterns objectively. We present the case of a man in his late twenties with drug-resistant focal epilepsy in whom we ascertained the effects of ASM withdrawal and a convulsive seizure on his touchscreen interactions.

Collagen VI: Role in synaptic transmission and seizure-related excitability.

Exp Neurol

October 2024

Epilepsy Centre, Department of Clinical Sciences, Lund University, 221 84 Lund, Sweden. Electronic address:

Collagen VI (Col-VI) is an extracellular matrix protein primarily known for its bridging role in connective tissues that has been suggested to play a neuroprotective role. In the present study we report increased mRNA and protein expression of Col-VI in the hippocampus and cortex at a late stage of epileptogenesis in a post-status epilepticus (SE) model of epilepsy and in brain tissue from patients with epilepsy. We further present a novel finding that exposure of mouse hippocampal slices to Col-VI augments paired-pulse facilitation in Schaffer collateral-CA1 excitatory synapses indicating decreased release probability of glutamate.

Mortality of Epilepsy in Chinese Populations: A Comprehensive Review.

Neuroepidemiology

July 2024

Department of Neurology, West of China Hospital, Sichuan University, Chengdu, China.

Background: Premature mortality is a significant part of the epilepsy burden and may vary across populations, especially between high-income and lower- and middle-income countries. People with epilepsy in China are approximately a fifth of the global population with epilepsy. Previous studies were unlikely to represent the situation in China due to limitations in design, methods, sample size, follow-up time, and other inherent population heterogeneity.

Article Synopsis
  • - The study focused on developing and preliminarily validating the Scale for Objective Diagnostic Components of Paroxysmal Events (STAMP), which categorizes seizure-like events, including epileptic seizures, functional seizures, and syncope.
  • - Researchers analyzed data from 200 patients referred to a Dutch epilepsy center, noting that 131 had epileptic seizures, 17 had functional seizures, and findings indicated significant diagnostic improvements through various event recordings during follow-up.
  • - The findings suggest that regular monitoring through STAMP can enhance diagnostic accuracy, although further validation of the scale is needed to fully establish its effectiveness.

The impact of ATP-binding cassette transporters in the diseased brain: Context matters.

Cell Rep Med

June 2024

Division of Pharmacology, The Netherlands Cancer Institute, Amsterdam, the Netherlands; Mouse Cancer Clinic, The Netherlands Cancer Institute, Amsterdam, the Netherlands. Electronic address:

ATP-binding cassette (ABC) transporters facilitate the movement of diverse molecules across cellular membranes, including those within the CNS. While most extensively studied in microvascular endothelial cells forming the blood-brain barrier (BBB), other CNS cell types also express these transporters. Importantly, disruptions in the CNS microenvironment during disease can alter transporter expression and function.

Effective reduction in seizure severity and prevention of a fatty liver by a novel low ratio ketogenic diet composition in the rapid kindling rat model of epileptogenesis.

Exp Neurol

September 2024

Amsterdam UMC location University of Amsterdam, Dept of (Neuro)pathology, Amsterdam Neuroscience, Meibergdreef 9, Amsterdam, the Netherlands; Stichting Epilepsie Instellingen Nederland (SEIN), Heemstede, the Netherlands.

Drug-resistant epilepsy patients may benefit from non-pharmacological therapies, such as the ketogenic diet (KD). However, its high fat content poses compliance challenges and metabolic risks. To mitigate this, we developed a novel KD composition with less fat and additional nutrients (citrate, nicotinamide riboside, and omega-3 fatty acids) for ketone-independent neuroprotection.

Department of Clinical Neurophysiology (location Zwolle & Heemstede), Stichting Epilepsie Instellingen Nederland (SEIN), Achterweg 5, Heemstede, SW 2103, the Netherlands; Department of Neurology, Leiden University Medical Centre, Albinusdreef 2, Leiden, RC 2300, the Netherlands; Department of Clinical & Experimental Epilepsy, UCL Queen Square Institute of Neurology, London WC1N 3BG, United Kingdom. Electronic address:

Purpose: Long-term ambulatory EEG recordings can improve the monitoring of absence epilepsy in children, but signal quality and increased review workload are a concern. We evaluated the feasibility of around-the-ears EEG arrays (cEEGrids) to capture 3-Hz short-lasting and ictal spike-and-wave discharges and assessed the performance of automated detection software in cEEGrids data. We compared patterns of bilateral synchronisation between short-lasting and ictal spike-and-wave discharges.

The classic ketogenic diet is an effective treatment option for drug-resistant epilepsy, but its high fat content challenges patient compliance. Optimizing liver ketone production guided by a method comparing substrates for their ketogenic potential may help to reduce the fat content of the diet without loss in ketosis induction. Here, we present a liver cell assay measuring the β-hydroxybutyrate (βHB) yield from fatty acid substrates.
